Role of S100A9 in tumor cell plasticity in breast cancer

Advanced breast cancer cases are considered incurable with current available therapies due to metastasis. One of the essential characteristics for metastasis is cell plasticity which allows cells to acquire new properties that lead to treatment resistance and relapse. One of the mechanisms towards cell plasticity is the acquisition of stem-like properties. Data from our group has shown that S100A9 could be involved in tumor cell plasticity through increasing stemness in breast cancer cells. By overexpressing S100A9 in the human non-tumorigenic epithelial cell line MCF10A and in the non-metastatic murine 67NR, we were able to assess the effect of the protein in stemness through the tumorspheres formation assay and ALDH+ population quantification. Moreover, a S100A9 knock-down was performed in the HER2+ basal breast cancer cell line HCC1954 to study the contrary effect. This ongoing project could be important in filling the gaps in the still challenging field of advanced breast cancer to be able to improve treatment and prognosis.
