American Journal of Transplantation<\/span><\/a>.<\/p>\nLa funci\u00f3 m\u00e9s coneguda dels limf\u00f2cits o c\u00e8l\u00b7lules T en el context de l\u2019al\u00b7loimmunitat \u00e9s la resposta efectora o inflamat\u00f2ria,<\/p>\n
que \u00e9s precisament la que provoca el rebuig en el trasplantament d\u2019\u00f2rgans. Per\u00f2 existeix un subtipus de c\u00e8l\u00b7lules T, les reguladores, que tenen com a funci\u00f3 justament la regulaci\u00f3 i modulaci\u00f3 d\u2019aquesta resposta immunol\u00f2gica efectora: s\u00f3n immunosupressores i per tant molt interessants en el camp dels trasplantaments.<\/p>\n
El fenotip m\u00e9s conegut d\u2019aquestes c\u00e8l\u00b7lules \u00e9s el que presenta el factor de transcripci\u00f3 FOXp3 per\u00f2 en humans, no sempre que les c\u00e8l\u00b7lules T presenten aquest factor tenen funci\u00f3 reguladora. Per aix\u00f2 els investigadors de l\u2019IDIBELL van decidir caracteritzar l\u2019epigen\u00e8tica d\u2019aquestes c\u00e8l\u00b7lules. \u00c9s a dir estudiar les modificacions qu\u00edmiques presents a l\u2019ADN del factor de transcripci\u00f3 FOXp3.<\/p>\n
L\u2019estudi s\u2019ha realitzat en dos grups de pacients de trasplantament renal: uns amb rebuig agut i uns altres amb rebuig subcl\u00ednic (el funcionament de l\u2019\u00f2rgan trasplantat \u00e9s bo per\u00f2 quan es fa una bi\u00f2psia es detecten alts nivells de c\u00e8l\u00b7lules T que suggereixen un rebuig).<\/p>\n
\u201cEn pacients amb rebuig agut, les c\u00e8l\u00b7lules T presentaven una modificaci\u00f3 epigen\u00e8tica (una metilaci\u00f3) en el factor FOXp3\u201d ha explicat l\u2019investigador de l\u2019IDIBELL i primer signant de l\u2019article Oriol Bestard, \u201cmentre que els pacients amb rebuig subcl\u00ednic i una bona funci\u00f3 renal no tenien aquest canvi en FOXp3, el factor preservava un estat demetilat\u201d.<\/p>\n
Aix\u00ed, aquest canvi epigen\u00e8tic en FOXp3 serveix com a biomarcador de pron\u00f2stic per un trasplantament de rony\u00f3. Si est\u00e0 demetilat sabem que s\u00f3n c\u00e8l\u00b7lules amb funci\u00f3 reguladora de la resposta immunol\u00f2gica que ens ajudaran a controlar el rebuig de l\u2019\u00f2rgan.<\/p>\n
L\u2019objectiu final de la recerca \u00e9s evitar que els pacients sotmesos a un transplantament renal hagin de sotmetre\u2019s tamb\u00e9 a un tractament immunosupressor amb els efectes adversos que aix\u00f2 suposa. \u201cA Europa\u201d ha explicat Bestard , \u201cs\u2019han iniciat estudis cl\u00ednics preliminars amb<\/p>\n
ter\u00e0pies cel\u00b7lulars per infondre c\u00e8l\u00b7lules T reguladores als pacients trasplantats, amb els resultats d\u2019aquests estudis la ter\u00e0pia seria m\u00e9s precisa perqu\u00e8 infondr\u00edem nom\u00e9s les c\u00e8l\u00b7lules T reguladores amb el factor FOXp3 demetilat, que s\u00f3n realment aquelles que tenen activitat immunoreguladora\u201d.<\/p>\n
