Un estudi de l’DIBELL demostra que la inhibici\u00f3 del metabolisme de la glucosa amb la mol\u00e8cula 2-deoxiglucosa (2-DG) indueix la mort cel\u00b7lular en un tipus de sarcoma infantil: el rabdomiosarcoma alveolar. Els resultats s\u2019han publicat a la revista Cancer Research<\/span><\/a>. <\/p>\n

Aquesta mol\u00e8cula \u00e9s molt similar a la que s\u2019utilitza en la tomografia per emissi\u00f3 de positrons (PET), una t\u00e8cnica d\u2019imatge que serveix per diagnosticar diversos tumors segons la seva taxa de consum de glucosa. Aix\u00f2 suggereix que podria ser utilitzada de manera immediata com a tractament d\u2019aquest tumor infantil agressiu. <\/p>\n

El rabdomiosarcoma \u00e9s el tumor de teixit tou m\u00e9s com\u00fa en nens i adolescents, i representa entre el 4 i el 5% dels tumors pedi\u00e0trics. Es presenta en dues formes: el rabdomiosarcoma embrional, el tipus m\u00e9s freq\u00fcent i menys agressiu, i el rabdomiosarcoma alveolar, de pitjor pron\u00f2stic. El tractament m\u00e9s utilitzat per aquest \u00faltim tipus de sarcoma \u00e9s la cirurgia. Els tractaments amb quimioter\u00e0pia no s\u00f3n efica\u00e7os i actualment la taxa de superviv\u00e8ncia despr\u00e8s de cinc anys del diagn\u00f2stic \u00e9s del 70%, la qual cosa indica que \u00e9s necessari el desenvolupament de tractaments m\u00e9s efica\u00e7os. <\/p>\n

Noves estrat\u00e8gies terap\u00e8utiques<\/strong> <\/p>\n

En aquest sentit, en els \u00faltims anys ha crescut l\u2019inter\u00e8s per estudiar el metabolisme tumoral com a possible diana terap\u00e8utica. Diverses vies metab\u00f2liques presenten diferents funcions en c\u00e8l\u00b7lules tumorals i en c\u00e8l\u00b7lules sanes. En concret, la glic\u00f2lisi (oxidaci\u00f3 de la glucosa per obtenir energia) augmenta en algunes c\u00e8l\u00b7lules tumorals. Aix\u00f2 les fa particularment sensibles a inhibidors de la glic\u00f2lisi com la 2-deoxiglucosa. <\/p>\n

L\u2019estudi, coordinat per la cap del grup de Regulaci\u00f3 de la Mort Cel\u00b7lular, Cristina Mu\u00f1oz-Pinedo, i el cap de grup de Sarcomes, \u00d2scar Mart\u00ednez-Tirado demostra que \u201cin vitro\u201d aquesta mol\u00e8cula inhibeix el metabolisme de la glucosa que la c\u00e8l\u00b7lula tumoral necessita, provocant la seva mort. Segons la investigadora Cristina Mu\u00f1oz-Pinedo aquesta mol\u00e8cula \u201cfrena el creixement de les c\u00e8l\u00b7lules tumorals, els provoca la mort i un percentatge d\u2019elles pateix una diferenciaci\u00f3 terminal, i presenten l\u2019aspecte de c\u00e8l\u00b7lules musculars sanes.\u201d <\/p>\n

Aquesta mol\u00e8cula tamb\u00e9 \u00e9s molt semblant a la que es fa servir en les t\u00e8cniques d\u2019imatge PET que s\u2019utilitzen per diagnosticar tumors que tenen un elevat metabolisme de la glucosa. Aix\u00f2, i el fet que ja s\u2019estan realitzant assaigs cl\u00ednics amb altres tumors, demostren que, a dosis altes, aquesta mol\u00e8cula \u00e9s poc t\u00f2xica i seria relativament f\u00e0cil poder-la fer servir en tractaments de rabdomiosarcoma alveolar. <\/p>\n

Mu\u00f1oz-Pinedo ha afegit que \u201cel fet de con\u00e8ixer el mecanisme cel\u00b7lular que provoca la mort d\u2019aquests c\u00e8l\u00b7lules tumorals permetr\u00e0, en el futur, afinar m\u00e9s en els tractaments i fer-los m\u00e9s personalitzats\u201d. <\/p>\n
