Prop del 10% dels c\u00e0ncers de mama es deuen a mutacions en els gens denominats BRCA1 i BRCA2. Tot i aix\u00f2, encara no es coneix totalment el mecanisme molecular que explica com l’alteraci\u00f3 d’aquests gens incrementa considerablement el risc de patir c\u00e0ncer. Un equip internacional coordinat per l’investigador de l’IDIBELL i de l’Institut Catal\u00e0 d’Oncologia<\/span><\/a> (ICO), Miquel \u00c0ngel Pujana, ha fet un pas important per a desvetllar-lo. Els investigadors han trobat una clau en la interacci\u00f3 entre el gen BRCA1 i altre denominat RHAMM (o HMMR), que suposa una via molecular desconeguda fins a ara. Els resultats de l’estudi s’han publicat a la revista PLoS Biology<\/span><\/a>.<\/p>\n

Els investigadors han demostrat que aquests dos gens (BRCA1 i RHAMM) vetllen perqu\u00e8 les c\u00e8l\u00b7lules del teixit mamari es desenvolupin i diferenci\u00efn amb normalitat. Si un d’aquests gens o els dos alhora presenten mutacions s’altera l’arquitectura i el desenvolupament normal del teixit i, d’aquesta forma, augmenta el risc que aparegui un tumor. \u201cEls portadors de mutacions de BRCA1 tenen un alt risc de patir c\u00e0ncer de mama\u201d, ha explicat l’investigador Miquel \u00c0ngel Pujana, \u201cper\u00f2 aquest risc es veu modificat, per exemple pel que fa a la seva edat d’aparici\u00f3, per variants gen\u00e8tiques en el gen RHAMM\u201d.<\/p>\n

Augmenta lleugerament el risc<\/strong><\/p>\n

Una variant gen\u00e8tica comuna al gen RHAMM augmenta lleugerament (aproximadament 1.1 vegades) el risc de patir c\u00e0ncer de mama en dones portadores de mutacions a BRCA1. \u201cPer tant, per si sola, aquesta variant no justifica una an\u00e0lisi gen\u00e8tica\u201d, explica Miquel \u00c0ngel Pujana. Per\u00f2 si unim aquesta alteraci\u00f3 a altres mutacions que tamb\u00e9 modifiquen lleugerament el risc, i actualment es coneixen una vintena, \u201cs\u00ed que pot variar de manera considerable el risc de patir c\u00e0ncer de mama en aquestes portadores. Depenent de la combinaci\u00f3 de les variants gen\u00e8tiques modificadores de risc que s’heretin es podrien veure augmentades o disminu\u00efdes les probabilitats de desenvolupar un c\u00e0ncer\u201d.<\/p>\n

La investigaci\u00f3 d’aquestes variants modificadores del risc s’orienta al desenvolupament de models gen\u00e8tics m\u00e9s precisos que puguin predir el risc de les portadores de BRCA1 i BRCA2 de patir c\u00e0ncer, i d’aquesta forma prendre les mesures de prevenci\u00f3 adequades. Aix\u00ed mateix les variants s’associen a diferents tipus de tumors, el que t\u00e9 implicacions importants en el pron\u00f2stic de les pacients.<\/p>\n
