Les alteracions en la formaci\u00f3 dels ribosomes (els elements de la c\u00e8l\u00b7lula on es fabriquen les prote\u00efnes) provoquen la inducci\u00f3 de la prote\u00efna p53 i la interrupci\u00f3 del cicle cel\u00b7lular. Aquest proc\u00e9s \u00e9s determinant per entendre processos biol\u00f2gics fonamentals i l’aparici\u00f3 de diverses malalties. Ara, cient\u00edfics de l’Institut de Recerca Biom\u00e8dica de Bellvitge (IDIBELL) han descobert que aquesta resposta s’aconsegueix de manera independent, segons quina unitat del ribosoma (40s i 60S) estigui afectada, mitjan\u00e7ant l’acci\u00f3 conjunta de dues prote\u00efnes del ribosoma. Els resultats de la investigaci\u00f3 s’han publicat en l’\u00faltim n\u00famero de la revista Genes and Development.<\/p>\n
La prote\u00efna p53, fabricada pel gen del mateix nom, \u00e9s un supressor tumoral en humans i en altres mam\u00edfers. La meitat dels tumors humans presenten mutacions en el gen p53 i l’alteraci\u00f3 de reguladors d’aquest gen s\u00f3n presents en molts altres c\u00e0ncers. No obstant aix\u00f2, p53 no \u00e9s nom\u00e9s un supressor tumoral sin\u00f3 que participa en altres processos cel\u00b7lulars com la longevitat, l’oxidaci\u00f3 mitocondrial i el metabolisme de la glucosa. L’activaci\u00f3 de la prote\u00efna p53 a causa de disfuncions en la formaci\u00f3 de ribosomes est\u00e0 relacionada amb diverses malalties minorit\u00e0ries, que estan associades amb mutacions en els components del nucli cel\u00b7lular.<\/p>\n
L’estudi realitzat per investigadors de l’IDIBELL mostra que les prote\u00efnes del ribosoma RPL5 i RPL11 cooperen per suprimir Hdm2, una altra prote\u00efna que regula la producci\u00f3 de p53 i la detenci\u00f3 del cicle cel\u00b7lular. Aquests resultats contradiuen el model vigent fins ara, que afirmava que altres prote\u00efnes del ribosoma podien aconseguir la mateixa resposta a RPL5 i RPL11.<\/p>\n
El coordinador de l’estudi, George Thomas, director del grup de recerca de Malalties Metab\u00f2liques i C\u00e0ncer de l’IDIBELL, afirma que el coneixement d’aquest mecanisme “pot tenir implicacions en la comprensi\u00f3 de processos fonamentals com la longevitat, les malalties metab\u00f2liques o el c\u00e0ncer i pot servir de base per al desenvolupament de nous f\u00e0rmacs”.<\/p>\n
El doctor Thomas s’ha incorporat recentment a l’IDIBELL, procedent de la Universitat de Cincinnati, i \u00e9s reconegut per la comunitat cient\u00edfica com un l\u00edder innovador en el camp de la transducci\u00f3 de senyals i el control del creixement cel\u00b7lular.<\/p>\n
L’estudi publicat a Genes and Development compta amb el finan\u00e7ament del Ministeri d’Economia i Competitivitat i de l’Institut de Salut Carles III.<\/p>\n
