Investigadors de l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) han creat una aplicaci\u00f3 inform\u00e0tica, anomenada MinerProt, que permet la classificaci\u00f3 de prote\u00efnes segons la seva funci\u00f3. El sistema, creat en col\u00b7laboraci\u00f3 amb investigadors de la Universitat Polit\u00e8cnica de Catalunya (UPC) i la Universitat Pompeu Fabra (UPF), resulta \u00fatil per prioritzar la import\u00e0ncia de certes prote\u00efnes. L’estudi ha estat publicat a la revista Molecular Biosystems.
\nL’equip, coordinat per la investigadora de l’IDIBELL \u00c0ngels Sierra, ha utilitzat aquesta eina juntament amb xarxes d’interacci\u00f3 prote\u00efna-prote\u00efna per crear perfils espec\u00edfics d’expressi\u00f3 de prote\u00efnes per a diferents met\u00e0stasi de c\u00e0ncer de mama, en cervell, pulm\u00f3, fetge i os, establint funcions priorit\u00e0ries en cadascuna d’elles. L’aproximaci\u00f3 funcional, basada en la selecci\u00f3 de les prote\u00efnes m\u00e9s representatives, pot servir per trobar marcadors per predir l’evoluci\u00f3 del proc\u00e9s cancer\u00f3s, aix\u00ed com per desenvolupar futures dianes terap\u00e8utiques.<\/p>\n

El principal objectiu de la recerca ha estat buscar un classificador funcional que ajudi a interpretar totes les dades referents a les prote\u00efnes relacionades amb les met\u00e0stasis del c\u00e0ncer de mama, donat el gran nombre que es troba diferencialment expressat i la seva complexitat d’estudi. El classificador indica les funcions cel\u00b7lulars que poden estar m\u00e9s representades en un determinat tipus de c\u00e8l\u00b7lula metast\u00e0tica. “Sobre aquestes funcions, el classificador ens indica quina prote\u00efna pot ser m\u00e9s important per a la funci\u00f3 que hem integrat”, explica Sierra, i continua: “partint de gens\/prote\u00efnes pr\u00e8viament identificades es classifiquen atenent al grau d’expressi\u00f3 diferencial (significaci\u00f3 estad\u00edstica) per despr\u00e9s organitzar-se en una xarxa d’interacci\u00f3 prote\u00efna-prote\u00efna. \u00c9s una forma pr\u00e0ctica de filtrar la informaci\u00f3, seleccionant les prote\u00efnes clau per definir funcions rellevants. Hem seguit un enfocament basat en discriminar les funcions de les met\u00e0stasis de c\u00e0ncer de mama atenent la seva organespecificitat que permetria el creixement cel\u00b7lular en microambients d’\u00f2rgans distants com os, cervell, fetge i pulm\u00f3”.<\/p>\n

Hi ha altres programes classificadors similars, per\u00f2 MinerProt prioritza la prote\u00efna m\u00e9s representativa de cada grup funcional, cosa que “ha ajudat a con\u00e8ixer els mecanismes moleculars caracter\u00edstics de la organespecificitat de les met\u00e0stasis”, explica la Dra Sierra.<\/p>\n

La met\u00e0stasi dep\u00e8n de m\u00faltiples i complexes interaccions. Nom\u00e9s les c\u00e8l\u00b7lules adaptades tenen la capacitat de sobreviure i cr\u00e9ixer en un \u00f2rgan distant. “Identificar interaccions letals entre prote\u00efna i prote\u00efna en les c\u00e8l\u00b7lules canceroses \u00e9s una via prometedora per a futurs desenvolupaments de dianes terap\u00e8utiques i de f\u00e0rmacs cada vegada m\u00e9s potents i selectius”, conclou Sierra.<\/p>\n

El treball, finan\u00e7at per la UE, l’Institut de Salut Carlos III, INCA, el Ministeri d’Educaci\u00f3 i Ci\u00e8ncia, i l’Associaci\u00f3 Espanyola Contra el C\u00e0ncer (AECC), es podria comercialitzar en un futur.<\/p>\n","protected":false},"excerpt":{"rendered":"
