El passat 30 d’octubre va tenir lloc el seminari IDIBELL extraordinari “Targeting signaling Pathways in breast cancer”, de Nancy E. Hynes, investigadora i l\u00edder del grup Mecanismes de C\u00e0ncer al Friedrich Miescher Institute for Biomedical Research, i organitzada per la Dra Sara Kozma. La Dra. Hynes va parlar de Memo, un nou enzim relacionat amb la met\u00e0stasi en el c\u00e0ncer de mama.<\/p>\n

“Preparem una an\u00e0lisi del transcriptoma per estudiar les vies en el c\u00e0ncer de mama”, va dir la investigadora Hynes. Durant el desenvolupament experimental es va veure que el gen Ret, que codifica un receptor de la tirosina cinasa, promou la migraci\u00f3, la superviv\u00e8ncia i la diferenciaci\u00f3 del sistema nervi\u00f3s. La inflamaci\u00f3 est\u00e0 relacionada amb la sobreregulaci\u00f3 de gens per l’activaci\u00f3 de Ret durant la ter\u00e0pia endocrina i l’augment de nivells d\u2019IL6 en resposta aquesta ter\u00e0pia. Els investigadors van trobar els nivells m\u00e9s alts d’IL-6 en les c\u00e8l\u00b7lules tractades amb fulvestrant. L\u2019IL-6 interactua amb Ret i, aix\u00ed mateix, promou la migraci\u00f3. La cinasa FAK s’activa amb Ret i IL6. La inhibici\u00f3 de FAK bloqueja l\u2019IL6, aix\u00ed com la senyalitzaci\u00f3 indu\u00efda de Ret. L\u2019IL6 MAB bloqueja la migraci\u00f3 de c\u00e8l\u00b7lules tractades GDNF i fulvestrant. L\u2019IL6 augmenta en resposta a agents endocrins.<\/p>\n

Models de c\u00e0ncer de mama: provant la funci\u00f3 Ret<\/strong><\/p>\n

“Ret t\u00e9 un paper en el creixement tumoral i la met\u00e0stasi in vivo<\/em>? Es van utilitzar dos models per respondre a aquesta pregunta. Amb el model de c\u00e0ncer de met\u00e0stasi J110 ER+\/Ret+ varem veure que AST487 redueix l’activitat de Ret i Akt. Mentre que amb el model de met\u00e0stasi de c\u00e0ncer J110 Ret- va ser molt similar”, va comentar la investigadora. La inhibici\u00f3 de Ret disminueix l’expressi\u00f3 d\u2019IL6 i Ret en tumors in vivo<\/em>. Els nivells de P-FAK s\u00f3n m\u00e9s baixos en els tumors tractats amb AST487. I bloquejant l’activitat de Ret s\u2019interfereix amb la uni\u00f3 Ret-IL6 i bloqueja la quinasa FAK, disminuint els nivells de Ret.<\/p>\n

En els models in vivo<\/em>, en els model ER+\/Ret +, de c\u00e0ncer de mama, Ret i ER cooperen per promoure la proliferaci\u00f3. Per als pacients, Ret es sobreexpressa en una s\u00e8rie de tumors de mama: ER+ i altres subgrups.<\/p>\n

Memo i met\u00e0stasi<\/strong><\/p>\n

El mediador de la mobilitat cel\u00b7lular ErbB2, conegut com Memo, indica els nivells d’expressi\u00f3 en tumors humans, i disminueix la invasi\u00f3 i la migraci\u00f3 de c\u00e8l\u00b7lules tumorals de mama. Memo es necessari en el c\u00e0ncer de pulm\u00f3, per\u00f2 no per al de mama.<\/p>\n

Memo es relaciona amb la met\u00e0stasi i interactua amb RhoA. Tamb\u00e9 es requereix per a una r\u00e0pida activaci\u00f3 de RhoA. Memo, que adopta una estructura de domini \u00fanic, t\u00e9 una activitat d\u2019oxidorreductasa i produeix super\u00f2xid. “Memo \u00e9s un enzim redox imprescindible per a la migraci\u00f3 en resposta a una \u00e0mplia gamma d’unions, i per a la met\u00e0stasi. L’activitat enzim\u00e0tica de Memo \u00e9s necess\u00e0ria per a la migraci\u00f3”, va concloure la investigadora.<\/p>\n","protected":false},"excerpt":{"rendered":"
