Investigadors de l’Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) han descrit una nova diana selectiva en la regeneraci\u00f3 muscular. Es tracta de l’associaci\u00f3 de les prote\u00efnes alfa-enolasa i plasmina. Les conclusions de la recerca podran servir per desenvolupar nous tractaments que permetin regenerar lesions o distr\u00f2fies musculars. L’estudi ha estat publicat a la revista PLOS ONE.<\/p>\n
El m\u00fascul esquel\u00e8tic t\u00e9 una gran capacitat de regeneraci\u00f3 despr\u00e9s d’una lesi\u00f3 o en malalties gen\u00e8tiques, com la distr\u00f2fia muscular de Duchenne, el trastorn neuromuscular m\u00e9s freq\u00fcent en nens. Aquesta patologia es deu a un defecte en el gen de la distrofina, la manca del qual causa inestabilitat de la membrana del m\u00fascul i condueix a la degeneraci\u00f3 de les fibres musculars.<\/p>\n
La regeneraci\u00f3 muscular implica una reestructuraci\u00f3 del teixit i requereix de la participaci\u00f3 d’enzims extracel\u00b7lulars, com la plasmina. La alfa-enolasa, un enzim present en el citoplasma de les c\u00e8l\u00b7lules, fa possible l’activitat de la plasmina, a la superf\u00edcie cel\u00b7lular, atorgant a la c\u00e8l\u00b7lula la capacitat de degradar el teixit circumdant.<\/p>\n
En aquest estudi, els investigadors de l’IDIBELL demostren que l’associaci\u00f3 de l’alfa-enolasa i la plasmina regula dos processos connectats en el m\u00fascul lesionat o amb distr\u00f2fia: d’una banda, l’atracci\u00f3 (reclutament) de c\u00e8l\u00b7lules immunit\u00e0ries per eliminar el teixit danyat i, d’altra banda, la formaci\u00f3 de nous teixits musculars a partir de c\u00e8l\u00b7lules mare. Els investigadors van observar al laboratori que aquestes c\u00e8l\u00b7lules mare perdien la capacitat d’activar i fusionar-se per formar les fibres del m\u00fascul esquel\u00e8tic quan aplicaven inhibidors espec\u00edfics de la uni\u00f3 alfa-enolasa\/plasmina.<\/p>\n
Els investigadors tamb\u00e9 van realitzar experiments en ratolins amb lesi\u00f3 muscular i distr\u00f2fia. En tractar-los amb els mateixos inhibidors, els animals presentaven un defecte important en la regeneraci\u00f3 muscular.<\/p>\n
“Aquests resultats demostren que la combinaci\u00f3 de l’alfa-enolasa i la plasmina \u00e9s necess\u00e0ria per a la restauraci\u00f3 del teixit muscular lesionat”, comenta la investigadora de l’IDIBELL Roser L\u00f3pez-Alemany, coordinadora de l’estudi.<\/p>\n
Recentment, un ampli metaan\u00e0lisi prote\u00f2mic ha identificat l’alfa-enolasa com la primera prote\u00efna expressada diferencialment, tant en patologies humanes com en el model de ratol\u00ed, la qual cosa suggereix que “podria ser considerada com un marcador d’estr\u00e8s patol\u00f2gic en un elevat nombre de malalties”, explica L\u00f3pez-Alemany.<\/p>\n
Aquest estudi indica que l’associaci\u00f3 alfa-enolasa\/plasmina \u00e9s una nova diana selectiva per les intervencions terap\u00e8utiques en patologies musculars ja que “demostra que l’alfa-enolasa \u00e9s responsable de l’activitat de la plasmina associada a la regeneraci\u00f3 muscular”, conclou l\u2019investigadora l’IDIBELL.<\/p>\n
