Un estudi de l’Institut de d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), liderat per Juli\u00e1n Cer\u00f3n, ha demostrat la doble funcionalitat d’una prote\u00efna, RSR-2, tant en la maquin\u00e0ria de transcripci\u00f3 de l’ADN com en el complex de maduraci\u00f3 de l’ARN (splicing) en el cuc C. elegans. \u00c9s la primera vegada que es confirma aquesta funci\u00f3 dual d’una prote\u00efna en un organisme multicel\u00b7lular viu.<\/p>\n

La prote\u00efna RSR-2 est\u00e0 conservada des de llevats fins a humans. Els resultats de l’estudi s’han publicat a la revista PLoS Genetics.<\/p>\n

Transcripci\u00f3 g\u00e8nica i ‘splicing’<\/strong><\/p>\n

L’ADN que codifica prote\u00efnes passa per diverses etapes per sintetitzar-les. En aquest proc\u00e9s hi ha dues maquin\u00e0ries: el complex de transcripci\u00f3 mitjan\u00e7ant el qual l’ADN passa a ARN i el complex de maduraci\u00f3 de l’ARN (splicing) que implica l’eliminaci\u00f3 de fragments de la seq\u00fc\u00e8ncia anomenats introns.<\/p>\n

Segons ha explicat l’investigador de l’IDIBELL Juli\u00e1n Cer\u00f3n, “s’ha demostrat en l\u00ednies cel\u00b7lulars i in vitro<\/em> que prote\u00efnes de les dues maquin\u00e0ries interaccionen f\u00edsicament, i que prote\u00efnes de la maquin\u00e0ria de transcripci\u00f3 s\u00f3n presents en la maquin\u00e0ria de splicing i viceversa. No obstant aix\u00f2, la interacci\u00f3 funcional de les dues maquin\u00e0ries no s’havia estudiat fins ara en un organisme multicel\u00b7lular viu “.<\/p>\n

Els investigadors van generar animals transg\u00e8nics i un antic\u00f2s contra la prote\u00efna RSR-2 per poder observar en quins llocs de la c\u00e8l\u00b7lula es trobava i van veure que “com esper\u00e0vem estava interactuant amb seq\u00fc\u00e8ncies d’ADN amb introns i per tant implicada en l’splicing, per\u00f2 tamb\u00e9 observem que estava associada a gens sense introns i per tant implicada tamb\u00e9 en el proc\u00e9s de transcripci\u00f3 g\u00e8nica\u201d ha explicat Cer\u00f3n.<\/p>\n

Interacci\u00f3 amb la via del retinoblastoma<\/strong><\/p>\n

En investigacions anteriors, Juli\u00e1n Cer\u00f3n, havia identificat una interacci\u00f3 gen\u00e8tica entre el gen RSR-2 i la via del retinoblastoma, una ruta gen\u00e8tica desregulada en la majoria de tumors humans, “per\u00f2 com RSR-2 pertanyia a la maquin\u00e0ria de splicing i el retinoblastoma participa en el remodelatge de la cromatina, en la maquin\u00e0ria de transcripci\u00f3, no sab\u00edem com interpretar aquesta interacci\u00f3 “.<\/p>\n

Demostrar que RSR-2 tamb\u00e9 funciona en la transcripci\u00f3 de l’ADN suposa per Cer\u00f3n “haver trobat la pe\u00e7a del puzle que ens faltava. Si retinoblastoma i RSR-2 es necessiten m\u00fatuament per funcionar, s’obre la porta a la modificaci\u00f3 de RSR-2 com a nova estrat\u00e8gia terap\u00e8utica contra el c\u00e0ncer “.<\/p>\n

Diana per Retinitis pigment\u00e0ria<\/strong><\/p>\n

La prote\u00efna RSR-2 tamb\u00e9 podria ser una nova diana terap\u00e8utica en ceguesa humana per un tipus espec\u00edfic de retinitis pigment\u00e0ria. Mutacions en el gen PRP8, present en la maquin\u00e0ria de splicing, provoquen un tipus de ceguesa progressiva coneguda com Retinitis pigment\u00e0ria. I l’equip de Cer\u00f3n ha demostrat que la RSR-2, igual que els seus hom\u00f2legs en llevats i en humans, interacciona amb el gen PRP8 obrint la porta a noves ter\u00e0pies a trav\u00e9s de la modificaci\u00f3 de RSR-2.<\/p>\n
