Investigadors del grup de Metabolisme i C\u00e0ncer de l’IDIBELL , de l’Institut Catal\u00e0 d’Oncologia (ICO) i de la Universitat de Cincinnati<\/span><\/a>, liderats per George Thomas, han descobert el paper de l’ARN ribosomal 5S en la formaci\u00f3 d’un complex que regula l’estabilitat de p53 . Normalment p53 evita que les c\u00e8l\u00b7lules sanes es converteixin en tumorals. Mant\u00e9 els nivells baixos i estables quan la c\u00e8l\u00b7lula funciona correctament i els fa augmentar quan hi ha danys a la c\u00e8l\u00b7lula.<\/p>\n

Els resultats de l’estudi s’han publicat en l’edici\u00f3 electr\u00f2nica de la revista Cell Reports<\/a>.<\/p>\n

Creixement cel\u00b7lular<\/strong><\/p>\n

El creixement cel\u00b7lular est\u00e0 relacionat amb la quantitat de prote\u00efnes que sintetitzen els ribosomes de la c\u00e8l\u00b7lula, la maquin\u00e0ria intracel\u00b7lular encarregada de traduir les mol\u00e8cules d’ARN missatger, provinent de l’ADN, en cadenes d’amino\u00e0cids, formant les prote\u00efnes). El mal funcionament en la formaci\u00f3 dels ribosomes est\u00e0 relacionat amb desordres associats al creixement cel\u00b7lular aberrant com l’an\u00e8mia i el c\u00e0ncer<\/p>\n

L’activaci\u00f3 de p53 indueix l’activaci\u00f3 d’un programa de mort cel\u00b7lular que prev\u00e9 que les c\u00e8l\u00b7lules que creixen de forma aberrant puguin desenvolupar un tumor. En condicions normals, es mant\u00e9 en nivells baixos per evitar danyar les c\u00e8l\u00b7lules sanes. La prote\u00efna principal en el manteniment de nivells baixos de p53 \u00e9s Hdm2, que en condicions de creixement cel\u00b7lular normal, degrada p53.<\/p>\n

Els ribosomes estan formats per dues subunitats denominades 40S i 60S. En la formaci\u00f3 de la subunitat 60S participen diverses mol\u00e8cules, incloent L5, L11 i 5S rRNA que formen un complex pre-ribosomal abans de ser incorporat a la subunitat 60S. El laboratori de George Thomas ha demostrat que quan hi ha un dany en els ribosomes, o quan la formaci\u00f3 de ribosomes est\u00e0 hiperactivada el complex pre-ribosomal L5\/L11\/5S rRNA es desvia de la seva ruta per formar ribosomes i s’uneix a Hdm2 bloquejant la seva activitat, permetent que augmentin els nivells de p53 i la inducci\u00f3 de la mort cel\u00b7lular.<\/p>\n

Anteriorment, l’equip de George Thomas ja havia demostrat que L5 i L11 regula Hdm2. Ara, Giulio Donatti, primer signant de l\u2019article ha descobert l’exist\u00e8ncia del aquest complex pre-ribosomal format per L5, L11 i tamb\u00e9 5S rRNA i el seu paper com a supressor de tumors. Aquests resultats apunten a un antic vincle evolutiu entre biog\u00e8nesi dels ribosomes i el c\u00e0ncer<\/p>\n

M\u00e9s del 50% dels tumors<\/strong><\/p>\n

George Thomas ha explicat que entendre com funciona i com es regula p53 \u00e9s molt important ja que “m\u00e9s del 50% dels tumors presenten mutacions en p53 o sobreexpressen Hdm2 o Hdm4, que bloquegen l’activitat de p53”. Thomas afegeix que “actualment estem treballant en el disseny d’un assaig cl\u00ednic, en colabroraci\u00f3n amb l’equip de Ramon Salazar, basat en activar el punt de control Hdm2-p53 per atacar les c\u00e8l\u00b7lules tumorals”.
