Investigadors del grup de recerca del Cicle Cel\u00b7lular de l’Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) dirigits per Ethel Queralt han publicat a la revista PLoS Genetics un article que aprofundeix en els mecanismes de regulaci\u00f3 de la mitosi, una fase del cicle cel\u00b7lular clau en la transmissi\u00f3 correcta de la informaci\u00f3 gen\u00e8tica de pares a fills .<\/p>\n

Les c\u00e8l\u00b7lules en tots els organismes creixen i es divideixen en dues c\u00e8l\u00b7lules filles mitjan\u00e7ant una successi\u00f3 ordenada d’esdeveniments que es denomina “cicle cel\u00b7lular”. B\u00e0sicament, les c\u00e8l\u00b7lules han de completar quatre processos principals durant el cicle cel\u00b7lular: cr\u00e9ixer (Fases G1 i G2), duplicar l\u2019ADN (Fase S), segregar els cromosomes (Fase M, mitosi ) i dividir-se (citocinesi) .<\/p>\n

En la fase S, o de replicaci\u00f3 de l\u2019ADN, es duplica el material gen\u00e8tic i posteriorment durant la fase M, o mitosi, les c\u00e8l\u00b7lules separen els cromosomes duplicats entre les dues c\u00e8l\u00b7lules filles. D’aquesta manera s’assegura la correcta her\u00e8ncia de la informaci\u00f3 gen\u00e8tica d’una generaci\u00f3 de c\u00e8l\u00b7lules a la seg\u00fcent. La transmissi\u00f3 de la informaci\u00f3 gen\u00e8tica (ADN) de pares a fills (o el que \u00e9s equivalent, d’unes c\u00e8l\u00b7lules a altres) \u00e9s una q\u00fcesti\u00f3 fonamental en la biologia .<\/p>\n

La aneuplo\u00efdia, \u00e9s a dir la falta o l’exc\u00e9s de cromosomes, \u00e9s una caracter\u00edstica present en gaireb\u00e9 tots els tipus de c\u00e0ncer hum\u00e0 i promou el desenvolupament de tumors. La regulaci\u00f3 de la mitosi \u00e9s particularment important per mantenir l’estabilitat cromos\u00f2mica: per exemple, les c\u00e8l\u00b7lules tumorals aneuploides apareixen com a conseq\u00fc\u00e8ncia de defectes en la segregaci\u00f3 de cromosomes. No obstant aix\u00f2 i malgrat la seva import\u00e0ncia, es coneix molt poc de la regulaci\u00f3 de la sortida de mitosi.<\/p>\n

En l’article publicat a la revista Plos Genetics es descriuen els darrers aven\u00e7os en els mecanismes de regulaci\u00f3 de la sortida de mitosi. La prote\u00efna denominada “separasa” \u00e9s un component clau per a la correcta segregaci\u00f3 dels cromosomes i per a la regulaci\u00f3 de la mitosi. En treballs previs el grup de Queralt va descriure per primera vegada la participaci\u00f3 de la prote\u00efna fosfatasa PP2A en la mitosi. Aquestes prote\u00efnes cooperen amb la separasa per assegurar la correcta her\u00e8ncia gen\u00e8tica d’unes c\u00e8l\u00b7lules a unes altres.<\/p>\n

“En el present treball” ha explicat Queralt “hem aprofundit en el mecanisme molecular pel qual aquesta prote\u00efna PP2A regula la mitosi i contribueix a que la segregaci\u00f3 dels cromosomes sigui la correcta mitjan\u00e7ant la identificaci\u00f3 de dos nous substrats, dos components de la ruta Hippo – tumor suppressor”.<\/p>\n

Segons ha explicat la investigadora “en els \u00faltims temps s’estan publicant estudis molt potents sobre l’\u00fas d’aquesta via com a diana terap\u00e8utica en c\u00e0ncer. No obstant aix\u00f2, al meu entendre, abans d’aix\u00f2 haur\u00edem de con\u00e8ixer millor els seus mecanismes moleculars”. En aquest sentit ha afirmat que “aquest treball contribueix a l’avan\u00e7 del coneixement de la ruta Hippo en la regulaci\u00f3 de la mitosi. I l’aplicaci\u00f3 d’aquests coneixements b\u00e0sics ajudar\u00e0 a desenvolupar tractaments m\u00e9s espec\u00edfics i directes contra els diferents tipus de c\u00e0ncer\u201d.<\/p>\n
