Quan parlem del material gen\u00e8tic normalment ens referim a l’ADN (\u00e0cid desoxiribonucleic) qu\u00e8 heretem dels nostres pares. Aquest ADN \u00e9s la f\u00e0brica que construeix una altra mol\u00e8cula similar anomenada ARN (\u00e0cid ribonucleic) que acaba produint les nostres prote\u00efnes, com l’hemoglobina o la insulina, que permeten la vida de les nostres c\u00e8l\u00b7lules. Per\u00f2 existeix un grup especial d’ARN anomenat no-codificant que t\u00e9 una funci\u00f3 m\u00e9s enigm\u00e0tica. El m\u00e9s conegut \u00e9s el microARN, mol\u00e8cules petit\u00edssimes que s’encarreguen d\u2019apagar o encendre gens del nostre genoma com si fossin interruptors de la llum.<\/p>\n

Avui, un article publicat a la prestigiosa revista Molecular Cell pel grup de Manel Esteller, director del Programa d’Epigen\u00e8tica i Biologia del C\u00e0ncer de l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), Investigador ICREA i professor de Gen\u00e8tica de la Universitat de Barcelona, proporciona una altra volta de rosca a aquest misteri. La investigaci\u00f3 demostra que existeix un segon tipus d’ARN no-codificant, anomenat ARN ultraconservat qu\u00e8 actua com a interruptor de l\u2019interruptor, \u00e9s a dir, controla l’activitat dels microARN.<\/p>\n

“Qui vigila al vigilant? Aquesta va ser una de les q\u00fcestions que ens vam plantejar a l’inici del treball” comenta Esteller. “Hav\u00edem trobat que unes mol\u00e8cules especials anomenades ARN ultraconservats deixaven de produir-se en els tumors humans i aix\u00f2 contribu\u00efa al creixement dels mateixos, per\u00f2 no sab\u00edem res sobre el seu mecanisme. Havien de tenir una funci\u00f3 important perqu\u00e8 aquestes mol\u00e8cules estan altament conservades a l\u2019evoluci\u00f3 i des dels pollastres als humans no hi ha cap variaci\u00f3. Ens vam adonar que s\u2019unien a l\u2019altra fam\u00edlia d\u2019ARN no-codificant, els microARNs, com un iman i impedien la seva funci\u00f3. \u00c9s a dir, s\u00f3n els policies d\u2019afers interns que supervisen els policies de batalla en les c\u00e8l\u00b7lules sanes. Si una c\u00e8l\u00b7lula deixa de produir l\u2019ARN ultraconservat, els microARN es descontrolen i s\u2019alteren centenars de gens que haurien de mantenir l\u2019equilibri cel\u00b7lular, contribuint aix\u00ed a la formaci\u00f3 de tumors\u201d conclou Esteller.<\/p>\n

Els resultats obtinguts pel grup d’Esteller al article de Molecular Cell suposen un pas molt important per comprendre la funci\u00f3 de l\u2019anomenat Genoma Fosc, terme col\u00b7loquial que defineix tot aquell material gen\u00e8tic que no produeix les cl\u00e0ssiques prote\u00efnes de la gen\u00e8tica m\u00e9s tradicional. El desxiframent dels codis d\u2019activitat cel\u00b7lular encriptats en aquestes seq\u00fc\u00e8ncies del nostre ADN suposa un dels reptes m\u00e9s apassionants de la biologia actual. Aquesta investigaci\u00f3 de frontera comen\u00e7a a donar els seus primers fruits en el camp de la recerca cl\u00ednica com en aquest cas en qu\u00e8 es relaciona l\u2019ARN ultraconservat amb el desenvolupament de c\u00e0ncer.<\/p>\n
