Investigadors de la UB <\/a><\/span>han sintetitzat un compost amb una nova estructura qu\u00edmica capa\u00e7 d\u2019induir in vitro la mort cel\u00b7lular en l\u00ednies cel\u00b7lulars tumorals. Segons un estudi publicat recentment a la revista cient\u00edfica Angewandte Chemie International Edition, la nova mol\u00e8cula est\u00e0 dirigida contra una nova diana molecular \u2014la prohibitina\u2014, una prote\u00efna implicada en diferents processos cel\u00b7lulars i patologies, especialment en processos tumorals. Aquests resultats obren la porta a noves estrat\u00e8gies per al tractament del c\u00e0ncer i el desenvolupament de nous f\u00e0rmacs.<\/p>\n

El treball s\u2019ha beneficiat de la col\u00b7laboraci\u00f3 interdisciplin\u00e0ria de diferents grups de la UB i ha estat coordinat per Rodolfo Lavilla, professor del Departament de Farmacologia i Qu\u00edmica Terap\u00e8utica de la UB; Joan Gil, catedr\u00e0tic del Departament de Ci\u00e8ncies Fisiol\u00f2giques II de la UB i investigador de l\u2019IDIBELL, i Fernando Albericio, catedr\u00e0tic del Departament de Qu\u00edmica Org\u00e0nica de la UB i investigador de l\u2019Institut d\u2019Investigaci\u00f3 Biom\u00e8dica (IRB Barcelona). En el treball tamb\u00e9 hi han participat altres grups de recerca, com el de la professora de la Facultat de Medicina de la UB i investigadora de l\u2019IDIBELL Isabel Fabregat, i el del professor Hiroshi Handa, de l\u2019Institut Tecnol\u00f2gic de T\u00f2quio (Jap\u00f3), la instituci\u00f3 on una de les primeres autores de l\u2019estudi, Alba P\u00e9rez Perarnau, ha fet una estada de recerca.<\/p>\n

Les trifluorotiazolines (TFT) s\u00f3n els nous compostos que han demostrat ser efectius en la inducci\u00f3 d\u2019apoptosi \u2014mort cel\u00b7lular programada\u2014 en l\u00ednies cel\u00b7lulars cancer\u00edgenes. Per estudiar el mecanisme pel qual aquestes mol\u00e8cules aconsegueixen aquest resultat, els investigadors han identificat les prote\u00efnes diana implicades. \u00abEl nostre estudi mostra que la mol\u00e8cula que hem sintetitzat aconsegueix inhibir la funci\u00f3 de les prohibitines, prote\u00efnes clau per determinar si les c\u00e8l\u00b7lules proliferen o entren en proc\u00e9s de mort cel\u00b7lular programada\u00bb, explica el professor Joan Gil, que tamb\u00e9 \u00e9s investigador principal del Grup de Recerca Consolidat en Apoptosi i C\u00e0ncer de la UB-IDIBELL i forma part de la Xarxa Tem\u00e0tica d\u2019Investigaci\u00f3 Cooperativa en C\u00e0ncer (RTICC) de l\u2019Institut de Salut Carlos III. Segons remarca Gil, \u00abaquesta pot ser la primera d\u2019una nova s\u00e8rie de compostos relacionats estructuralment i que encara podrien ser millors inhibidors de la prohibitina\u00bb.
\n<\/strong> Un compost assequible i estable<\/strong><\/p>\n

Existeixen productes naturals que s\u2019adhereixen a les prohibitines. No obstant aix\u00f2, la complexitat i la complicada disponibilitat d\u2019aquests compostos dificulten que es puguin usar en estudis biol\u00f2gics i terap\u00e8utics. \u00abLa preparaci\u00f3 de les TFT \u00e9s directa, i aix\u00f2 les converteix en compostos m\u00e9s assequibles i selectius, \u00fatils tant en qu\u00edmica m\u00e8dica com en biologia b\u00e0sica, per la facilitat de preparaci\u00f3. Tamb\u00e9 s\u00f3n estables i constitueixen una nova estructura qu\u00edmica (scaffold) in\u00e8dita en qu\u00edmica m\u00e8dica, susceptible de ser explorada en altres \u00e0mbits terap\u00e8utics\u00bb, explica el professor Rodolfo Lavilla, que tamb\u00e9 \u00e9s investigador principal del Grup de Recerca de S\u00edntesi i Reactivitat d\u2019Heterocicles Bioactius del Parc Cient\u00edfic de Barcelona (PCB).<\/p>\n

La s\u00edntesi de la nova mol\u00e8cula ha estat especialment complexa pel que fa a \u00abl\u2019elucidaci\u00f3 de l\u2019estructura, la racionalitzaci\u00f3 del mecanisme i la preparaci\u00f3 d\u2019aquests compostos polifuncionals, especialment la sonda molecular utilitzada en la identificaci\u00f3 de la diana molecular\u00bb, detalla Lavilla. En aquests punts, ha estat clau el paper de la investigadora Sara Preciado (UB-PCB), que, en el marc de la seva tesi, ha desenvolupat i perfeccionat els aspectes principals de la recerca qu\u00edmica.<\/p>\n

Segons Lavilla, \u00abel principal avantatge d\u2019aquesta mol\u00e8cula \u00e9s que es tracta d\u2019un compost que es pot sintetitzar, la qual cosa en facilita l\u2019obtenci\u00f3, tant des del punt de vista metodol\u00f2gic com econ\u00f2mic\u00bb. \u00abA m\u00e9s \u2014subratlla Joan Gil\u2014, les nostres dades indiquen que la seva acci\u00f3 com a inductora de la mort de les c\u00e8l\u00b7lules est\u00e0 vehiculada per la inhibici\u00f3 de prohibitina i \u00e9s independent de la prote\u00efna p53, que est\u00e0 mutada o no s\u2019expressa en almenys el 50 % dels tumors humans\u00bb.<\/p>\n

L\u2019equip de la UB ha continuat aquesta l\u00ednia de recerca en col\u00b7laboraci\u00f3 amb Thomas Langer, especialista en el camp de les prohibitines de la Universitat de Col\u00f2nia. \u00abEls primers resultats d\u2019aquesta col\u00b7laboraci\u00f3 han confirmat que el nou compost no indueix la mort de les c\u00e8l\u00b7lules que no expressen prohibitina\u00bb, conclou Joan Gil.<\/p>\n
