Actualment, hi ha m\u00e9s de 350 milions de diab\u00e8tics tipus 2 i d’acord amb l’Organitzaci\u00f3 Mundial de la Salut (OMS) per a l’any 2030 ser\u00e0 la setena causa de mort a tot el m\u00f3n. La marca m\u00e9s caracter\u00edstica de la diabetis mellitus tipus 2 \u00e9s la resist\u00e8ncia a la insulina que es veu inicialment compensada per un augment de la mida de les c\u00e8l\u00b7lules beta pancre\u00e0tiques, (encarregades de produir la insulina) fins que aquestes es col\u00b7lapsen i moren i \u00e9s quan la malaltia entra en un estat avan\u00e7at. <\/p>\n
Investigadors del Laboratori de Metabolisme i C\u00e0ncer de l\u2019IDIBELL liderats per Sara Kozma han demostrat en models animals que la inhibici\u00f3 de la prote\u00efna S6K1 pot ser un potencial tractament per a la diabetis tipus 2. <\/p>\n
En un estudi publicat aquesta setmana a la revista Journal Clinic of Investigation els investigadors han demostrat que els animals que no tenen S6K1, una prote\u00efna quinasa, s\u00f3n m\u00e9s sensibles a la insulina, per tant en necessiten menys i no desenvolupen la diabetis. En estudis anteriors el grup de Kozma havia observat que els ratolins deficients en S6K1 eren m\u00e9s petits i que presentaven uns nivells circulants d\u2019insulina per sota del normal. \u201cVam demostrar que les c\u00e8l\u00b7lules beta d\u2019aquests animals eren m\u00e9s petites i produ\u00efen menys insulina\u201d explica la investigadora. \u201cTamb\u00e9 van demostrar que en abs\u00e8ncia de S6K1, els teixits perif\u00e8rics de l\u2019animal es feien m\u00e9s sensibles a la insulina<\/p>\n
i no desenvolupaven la malaltia, fins i tot quan se\u2019ls sotmetia a una dieta rica en greixos\u201d. <\/p>\n
En aquest estudi, el grup de Kozma ha utilitzat la t\u00e8cnica de rescat embrionari per entendre per qu\u00e8 les c\u00e8l\u00b7lules beta d\u2019aquests ratolins eren m\u00e9s petites. Van implantar c\u00e9l.lules mare embrion\u00e0ries deficients en S6K1 en placentes de ratolins normals i van veure que tot i que l\u2019embri\u00f3 assolia una mida normal, les seves c\u00e8l\u00b7lules beta continuaven sent petites. Per tant, la mida de les c\u00e8l\u00b7lules beta \u00e9s independent del desenvolupament del ratol\u00ed a l\u2019\u00fater i \u00e9s l\u2019abs\u00e8ncia de S6K1 als teixits perif\u00e8rics la que permet l\u2019augment de la sensibilitat a la insulina de l\u2019animal. De forma que, els inhibidors d\u2019aquesta prote\u00efna poden servir com a potencials sensibilitzadors a la insulina per protegir contra la seva insensibilitat i tractar la diabetis tipus 2.
