Investigadors del grup de vies de senyalitzaci\u00f3 i angiog\u00e8nesi de l\u2019Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) liderats per Mariona Graupera han demostrat<\/p>\n
que la prote\u00efna PTEN \u00e9s clau per evitar la proliferaci\u00f3 descontrolada de les c\u00e8l\u00b7lules que formen els vasos sanguinis en el proc\u00e9s d\u2019angiog\u00e8nesi. Els resultats de l\u2019estudi es publiquen avui a la revista Nature Communications. <\/p>\n
El creixement de vasos sanguinis per brots (sprouting en angl\u00e8s) \u00e9s un mecanisme central de la formaci\u00f3 de la vasculatura i comen\u00e7a per l\u2019especialitzaci\u00f3 de c\u00e8l\u00b7lules endotelials en diversos tipus de c\u00e8l\u00b7lules. D\u2019una banda, les c\u00e8l\u00b7lules tip (punta en catal\u00e0) que guien el sentit del creixement i que gaireb\u00e9 no proliferen i d\u2019altra, les c\u00e8l\u00b7lules stalk (tija en catal\u00e0) que creixen darrera per anar formant el lumen dels nous vasos sanguinis. <\/p>\n
Estudis anteriors havien demostrat que la ruta de senyalitzaci\u00f3 de Notch est\u00e0 implicada en el control de les c\u00e8l\u00b7lules stalk bloquejant la seva proliferaci\u00f3. El treball de l\u2019equip de Mariona Graupera demostra com Notch activa PTEN i aquesta promou la parada del cicle cel\u00b7lular i per tant evita la proliferaci\u00f3 de les c\u00e8l\u00b7lules. Els investigadors han demostrat tant en cultius cel\u00b7lulars com en ratolins que en abs\u00e8ncia de PTEN, tot i l\u2019activaci\u00f3 de Notch, no s\u2019atura la proliferaci\u00f3 de les c\u00e8l\u00b7lules stalk derivant en la formaci\u00f3 d\u2019una vasculatura aberrant. En els \u00faltims anys s\u2019ha vist que les ter\u00e0pies antiangiog\u00e8niques s\u00f3n de curt recorregut i a llarg termini poden ser perjudicials. Les aproximacions actuals es dirigeixen a normalitzar la vasculatura de manera que tot i que els nutrients i l\u2019ox\u00edgen arribin a les c\u00e8l\u00b7lules tumorals tamb\u00e9 ho facin ela agents quimioterap\u00e8utics. <\/p>\n \u201cEn aquest sentit, es pot utilitzar PTEN per normalitzar la vasculatura i mantenir la quiesc\u00e8ncia, l\u2019equilibri en la formaci\u00f3 des vasos sanguinis\u201d conclou la investigadora. <\/p>\n Helena Serra, I\u00f1igo Chivite, Ana Angulo-Urarte, Adriana Soler, James D.
