Investigadors del grup de Gen\u00e8tica Molecular Humana de l\u2019IDIBELL, liderats per Merc\u00e8 P\u00e9rez, de la Universitat Aut\u00f2noma de Barcelona i de la Cl\u00ednica CIMA de Navarra, han identificat dos composts org\u00e0nics petits que competeixen amb els NFATc per unir-se al mateix lloc de la prote\u00efna calcineurina i que podrien tenir un potencial efecte immunosupressor <\/p>\n
L\u2019activaci\u00f3 de la resposta immune, causada per l\u2019activaci\u00f3 de limf\u00f2cits T, \u00e9s el resultat final d\u2019un conjunt de respostes mitjan\u00e7ades per diverses vies de senyalitzaci\u00f3, com ara la de la calcineurina cap <\/p>\n
els factors de transcripci\u00f3 NFATc. L\u2019estimulaci\u00f3 del receptor de c\u00e8l\u00b7lules T produeix un increment notable de la concentraci\u00f3 de calci dins la c\u00e8l\u00b7lula, fent que s\u2019activi la fosfatasa calcineurina. Un cop activa, la calcineurina desfosforila els seus substrats, inclosos els factors de transcripci\u00f3 NFATc i, en conseq\u00fc\u00e8ncia, NFATc transloca al n\u00facleo on activa l\u2019expressi\u00f3 de gens i la producci\u00f3 de citoquines i quimioquines, que condueix a l\u2019activaci\u00f3 de les c\u00e8l\u00b7lules T i de la resposta immune <\/p>\n
Els investigadors han observat que aquests compostos eviten que els NFATcs s\u2019activin i, per tant, no indueixen la expressi\u00f3 de gens de citoquines i quimioquines com ara la interleuquina-2, interferon gamma i CSF2, mantenint els limfocits T CD4+ humans inactius i no activant-se, per tant, la resposta immune. <\/p>\n
Merc\u00e8 P\u00e9rez ha destacat que \u201cen contraposici\u00f3 als immunosupressors actuals ciclosporina A i FK506 (o tacrolimus) que inhibeixen l\u2019activitat de calcineurina cap a tots els seus substrats, aquests dos composts no afecten la activitat fosfatasa de calcineurina cap a altres substrats diferents als NFATcs\u201d. Per tant, els investigadors creuen que<\/p>\n
els efectes secundaris produ\u00efts per aquests dos compostos seran molt inferior a la dels immunosupressors actuals. <\/p>\n
Aquests resultats poden millorar la toler\u00e0ncia cl\u00ednica de futurs immunosupressors per pacients amb ter\u00e0pia immunosupressora com en el trasplantament d\u2019\u00f2rgans i el tractament de malalties autoimmunes. A m\u00e9s, aquests composts poden ser tamb\u00e9 \u00fatils en el tractament de malalties on la via de la calcineurina-NFATc est\u00e0 desregulada com ara el c\u00e0ncer. <\/p>\n
