Investigadors del grup d’Envelliment i C\u00e0ncer, liderat per Purificaci\u00f3n Mu\u00f1oz, de l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), han demostrat que les caracter\u00edstiques de les c\u00e8l\u00b7lules mare del c\u00e0ncer canvien durant la progressi\u00f3 dels carcinomes escatosos de pell, generant tumors m\u00e9s agressius i amb major capacitat metast\u00e0tica. Aquests resultats han estat recentment publicats a la revista Cancer Research.<\/p>\n

Els carcinomes escatosos s\u00f3n el segon tipus de tumor de pell m\u00e9s freq\u00fcent. Un alt percentatge d’aquests tumors s\u00f3n eliminats mitjan\u00e7ant resecci\u00f3 quir\u00fargica. No obstant aix\u00f2, alguns d’aquests tumors d’estadis avan\u00e7ats mostren recurr\u00e8ncia, creixement agressiu i una alta predisposici\u00f3 per generar met\u00e0stasi.<\/p>\n

Estudis anteriors van demostrar que en la majoria dels tumors hi ha c\u00e8l\u00b7lules cancer\u00edgenes que mostren propietats similars a les c\u00e8l\u00b7lules mare adultes. Aquesta poblaci\u00f3 de c\u00e8l\u00b7lules cancer\u00edgenes, anomenades c\u00e8l\u00b7lules mare tumorals, s\u00f3n responsables del creixement tumoral i de la generaci\u00f3 de met\u00e0stasi. Per tant, \u00e9s important con\u00e8ixer els mecanismes que regulen la proliferaci\u00f3, superviv\u00e8ncia i disseminaci\u00f3 de les c\u00e8l\u00b7lules mare tumorals per tal de bloquejar el creixement tumoral i la generaci\u00f3 de met\u00e0stasi.<\/p>\n

Els investigadors del grup d’Envelliment i C\u00e0ncer han generat models de ratol\u00ed que permeten caracteritzar la funci\u00f3 i regulaci\u00f3 de les c\u00e8l\u00b7lules mare tumorals durant la progressi\u00f3 dels carcinomes escatosos de pell, des d’estadis inicials, on aquestes c\u00e8l\u00b7lules conserven caracter\u00edstiques de diferenciaci\u00f3 epitelial i una baixa capacitat metast\u00e0tica, fins carcinomes altament agressius, indiferenciats i amb una elevada capacitat per generar met\u00e0stasi. Els seus resultats demostren que la freq\u00fc\u00e8ncia de les c\u00e8l\u00b7lules mare tumorals augmenta dram\u00e0ticament en els carcinomes avan\u00e7ats, mostrant a m\u00e9s una forta inducci\u00f3 del programa de transici\u00f3 epiteli-mes\u00e8nquima, el qual afavoreix la disseminaci\u00f3 d’aquestes c\u00e8l\u00b7lules a altres teixits i, en conseq\u00fc\u00e8ncia, la generaci\u00f3 de met\u00e0stasi. A m\u00e9s, els mecanismes que regulen la proliferaci\u00f3, superviv\u00e8ncia i disseminaci\u00f3 de les c\u00e8l\u00b7lules mare tumorals canvien durant la progressi\u00f3 tumoral. Aix\u00ed, mentre que la via dependent d’EGFR juga un paper important promovent el creixement d’aquestes c\u00e8l\u00b7lules en carcinomes escatosos primerencs, aquesta via est\u00e0 atenuada en carcinomes avan\u00e7ats i s’indueixen les vies que depenen de FGFR1 i PDGFRA, responsables de promoure respectivament un creixement tumoral agressiu i la met\u00e0stasi. L’an\u00e0lisi de mostres de pacients, realitzat en col\u00b7laboraci\u00f3 amb el Servei de Cirurgia Pl\u00e0stica i la Unitat d’Anatomia Patol\u00f2gica de l’Hospital de Bellvitge (IDIBELL), va demostrar que de forma similar al descrit en tumors de ratol\u00ed, els carcinomes escatosos recurrents presenten una forta inducci\u00f3 del programa de transici\u00f3 epiteli-mes\u00e8nquima i de l’expressi\u00f3 dels receptors i lligands de les vies de PDGFR i FGFR1.<\/p>\n

At\u00e8s que hi ha inhibidors farmacol\u00f2gics que inhibeixen l’activitat d’aquests receptors, aquests estudis ofereixen la base per testar noves ter\u00e0pies dirigides al tractament dels carcinomes de pell avan\u00e7ats, que podrien millorar la qualitat de vida i superviv\u00e8ncia dels pacients. <\/p>\n
