La informaci\u00f3 gen\u00e8tica del pare es transmet a la descend\u00e8ncia gr\u00e0cies a unes c\u00e8l\u00b7lules especialitzades, molt adaptades a la seva funci\u00f3: els espermatozoides. El desenvolupament d’aquestes c\u00e8l\u00b7lules en el testicle (espermatog\u00e8nesi) \u00e9s un proc\u00e9s molt regulat en el qual participen uns 2500 gens diferents de manera precisa i coordinada. <\/p>\n

Els microRNAs (miRNAs) s\u00f3n mol\u00e8cules reguladores de l’expressi\u00f3 dels gens i estan implicats en una \u00e0mplia varietat de processos del desenvolupament en vertebrats. El paper dels miRNAs en el desenvolupament de la c\u00e8l\u00b7lula germinal masculina \u00e9s rellevant: espec\u00edficament interv\u00e9 en la diferenciaci\u00f3 del testicle en l’embri\u00f3, aix\u00ed com en el desenvolupament de la l\u00ednia germinal i la producci\u00f3 dels espermatozoides. Investigadors del grup de Gen\u00e8tica Molecular Humana de l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), en concret de la l\u00ednia de Gen\u00e8tica Molecular de la Fertilitat Masculina liderada per la Dra Sara Larriba, en col\u00b7laboraci\u00f3 amb el Servei d’Andrologia de la Fundaci\u00f3 Puigvert, han identificat un perfil de miRNA alterat en les c\u00e8l\u00b7lules germinals en desenvolupament (i que persisteix en els espermatozoides), en individus inf\u00e8rtils amb baixa producci\u00f3 esperm\u00e0tica. La troballa s’ha publicat a la prestigiosa revista cient\u00edfica internacional de lliure acc\u00e9s Scientific Reports. <\/p>\n

“En publicacions anteriors hav\u00edem demostrat que la baixa efici\u00e8ncia de l’espermatog\u00e8nesi en individus inf\u00e8rtils s’acompanya de patrons d’expressi\u00f3 g\u00e8nica alterats en les c\u00e8l\u00b7lules germinals en desenvolupament, que al seu torn contribueixen a una reducci\u00f3 en la producci\u00f3 d’esperma. En aquest context, \u00e9s important determinar com es comporten mecanismes de regulaci\u00f3 de l’expressi\u00f3 de gens com els miRNAs” comenta la Dra. Larriba. En aquest treball s’han avaluat els nivells de 623 miRNAs madurs en mostres testiculars histol\u00f2gicament ben definides amb alteraci\u00f3 de l’espermatog\u00e8nesi en les diferents etapes de la l\u00ednia germinal. Els resultats suggereixen que el contingut cel\u00b7lular de miRNAs en espermatozoides dep\u00e8n de l’efici\u00e8ncia de l’espermatog\u00e8nesi, sent aquestes difer\u00e8ncies m\u00e9s notables com m\u00e9s immadur \u00e9s l’estadi de la l\u00ednia germinal on es bloqueja l’espermatog\u00e8nesi. Els espermatozoides que aconsegueixen diferenciar-se en espermatog\u00e8nesis deficients retenen el patr\u00f3 de miRNA desregulat de les c\u00e8l\u00b7lules germinals immadures. Aquesta signatura molecular alterada de miRNA t\u00e9, molt probablement, implicacions funcionals per al g\u00e0meta mascul\u00ed, i fins i tot en les primeres etapes de l’embriog\u00e8nesi despr\u00e9s de la fecundaci\u00f3 de l’o\u00f2cit. Aquests resultats ofereixen una nova visi\u00f3 molecular dels mecanismes que regulen i controlen la fertilitat masculina, i proporcionen elements per al desenvolupament de noves eines terap\u00e8utiques. <\/p>\n
