<\/strong> Un equip d\u2019investigadors ha dissenyat un m\u00e8tode computacional per con\u00e8ixer la progressi\u00f3 del c\u00e0ncer colorectal, un aven\u00e7 cient\u00edfic que pot obrir noves perspectives al coneixement dels factors que potencien aquesta patologia i a la selecci\u00f3 de ter\u00e0pies efectives. En el nou estudi, publicat a la revista Proceedings of the National Academy of Sciences<\/em> (PNAS), participen els experts Victor Moreno<\/strong>, de la Facultat de Medicina i Ci\u00e8ncies de la Salut de la UB, l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) i l\u2019Institut Catal\u00e0 d\u2019Oncologia (ICO), i Rebeca Sanz-Pamplona <\/strong>(IDIBELL-ICO), entre altres investigadors de tot l\u2019\u00e0mbit internacional.<\/p>\n

Quines condicions faciliten el creixement dels tumors?<\/strong> <\/p>\n

El c\u00e0ncer de c\u00f2lon t\u00e9 una freq\u00fc\u00e8ncia similar en homes i en dones, mentre que el de recte es m\u00e9s freq\u00fcent en els homes. Aproximadament, 13.000 homes i 14.000 dones conviuen amb un diagn\u00f2stic de c\u00e0ncer colorectal a Catalunya. <\/p>\n

Tal com explica el catedr\u00e0tic V\u00edctor Moreno, del Departament de Ci\u00e8ncies Cl\u00edniques de la UB, \u00abcom a la majoria de tumors, el colorectal es caracteritza per grans alteracions cromos\u00f2miques (delecions, amplificacions) i mutacions puntuals. Aquests factors a nivell d’ADN despr\u00e9s generen importants alteracions a nivell d’ARN i metabolisme de les c\u00e8l\u00b7lules tumorals\u00bb. <\/p>\n

Segons el primer autor de l\u2019article, el professor Giulio Caravagna de la Universitat d\u2019Edimburg, \u00abproposem un protocol bioinform\u00e0tic per detectar regularitats que s\u00f3n comunes en l\u2019origen i el desenvolupament del tumor. Aquest protocol podria ser un progr\u00e9s clau per entendre una malaltia que es caracteritza per un nombre molt baix d\u2019alteracions gen\u00f2miques comuns en diferents pacients\u00bb.<\/p>\n

Per a l\u2019expert Bud Mishra, de la Universitat de Nova York, \u00abel model ajuda a predir com el genoma del tumor anir\u00e0 canviat amb el temps\u00bb. <\/p>\n

Tradicionalment, es considerava que el c\u00e0ncer s\u2019iniciava amb una c\u00e8l\u00b7lula rebel, i s\u2019estenia, en part, per una combinaci\u00f3 d\u2019interaccions gen\u00e8tiques aut\u00f2nomes : mutacions en oncogens, amb la capacitat potencial de provocar c\u00e0ncer, i el frac\u00e0s dels genes supressors de tumors que els controlen.<\/p>\n

No obstant aix\u00f2, la disponibilitat de t\u00e8cniques se seq\u00fcenciaci\u00f3 avan\u00e7ada ha perfilat un escenari m\u00e9s complex sobre la progressi\u00f3 del c\u00e0ncer. De forma espec\u00edfica, tot indica que la interacci\u00f3 entre les c\u00e8l\u00b7lules en una poblaci\u00f3 tumoral \u00e9s m\u00e9s rellevant del que es pensava fins ara, ja que els tumors s\u00f3n m\u00e9s heterogenis a nivell cel\u00b7lular del que es creia. <\/p>\n

PiCnIc<\/em><\/strong>: nova eina per con\u00e8ixer la progressi\u00f3 del c\u00e0ncer<\/strong> <\/p>\n

<\/strong> L\u2019equip cient\u00edfic ha captat aquestes interaccions mitjan\u00e7ant un model que vol generar una imatge cel\u00b7lular de la propagaci\u00f3 del c\u00e0ncer colorrectal.<\/p>\n

Per aix\u00f2, s\u2019ha desenvolupat un sistema de modelitzaci\u00f3 (Pipeline for Cancer Inference, o PiCnIc<\/em>)<\/strong> que fa \u00fas de dades de seq\u00fcenciaci\u00f3 gen\u00e8tica per elaborar prediccions sobre la causalitat. <\/p>\n

PiCnIc, en particular, t\u00e9 en consideraci\u00f3 la funci\u00f3 de les mutacions conductores (driver) que estimulen la progressi\u00f3 del c\u00e0ncer, a m\u00e9s dels altres fen\u00f2mens com ara la influ\u00e8ncia d\u2019aquestes mutacions sobre altres factors conductors.<\/p>\n

Per constatar la viabilitat del models, l\u2019equip cient\u00edfic ha comparat les prediccions amb els coneixements disponibles sobre la din\u00e0mica del creixement del c\u00e0ncer colorrectal. Segons els resultats,<\/p>\n

PiCnIc \u00e9s una eina efica\u00e7 per reflectir els coneixements de la investigaci\u00f3 biom\u00e8dica sobre la progressi\u00f3 de la malaltia.<\/p>\n

\u00abAquesta nova metodologia \u00e9s novedosa perqu\u00e8 incorpora un model probabil\u00edstic que infereix l’ordre dels successos (mutacions i alteracions en numero de c\u00f2pies)\u00bb, detalla el catedr\u00e0tic V\u00edctor Moreno (UB-IDIBELL-ICO). <\/p>\n
