Investigadors de l’Institut de Recerca Biom\u00e8dica de Bellvitge (IDIBELL), liderats per la Dra. Eva Gonz\u00e1lez-Su\u00e1rez, han demostrat que la inhibici\u00f3 farmacol\u00f2gica i gen\u00e8tica de la via de senyalitzaci\u00f3 de RANK \/ RANKL condueix a una reducci\u00f3 significativa de recidives i met\u00e0stasi de c\u00e0ncer de mama en un model animal de ratol\u00ed. Les seves troballes, publicades a Cancer Research<\/em>, suggereixen que els inhibidors RANK, que s’utilitzen actualment en els pacients amb osteoporosi i met\u00e0stasi \u00f2ssia, poden tenir potencial per al tractament no quir\u00fargic del c\u00e0ncer de mama. <\/p>\n

Les c\u00e8l\u00b7lules precursores de c\u00e0ncer o c\u00e8l\u00b7lules mare cancer\u00edgenes (CMC) expressen alts nivells de la prote\u00efna RANK. “De fet, la mortalitat en el c\u00e0ncer de mama es deu principalment a les CMC supervivents a tractaments, que s\u00f3n responsables de la recurr\u00e8ncia del tumor i la met\u00e0stasi”, explica la Dra. Gonz\u00e1lez-Su\u00e1rez. “Altres estudis d’aquest i altres grups de recerca havien demostrat que la inhibici\u00f3 de la via de RANK podria prevenir el c\u00e0ncer de mama, per\u00f2 fins ara ning\u00fa havia demostrat el potencial dels inhibidors de la via de RANK en el tractament de la malaltia.” En el seu darrer treball, els investigadors demostren que la inhibici\u00f3 de la via de senyalitzaci\u00f3 RANK \/ RANKL podria emprar-se potencialment com a nova ter\u00e0pia en c\u00e0ncer de mama; en paraules de la investigadora de l’IDIBELL, “aquesta inhibici\u00f3 no redueix el creixement del tumor, per\u00f2 en promou la diferenciaci\u00f3, cosa que redueix la poblaci\u00f3 de CMC i dificulta la met\u00e0stasi, millorant el pron\u00f2stic.” <\/p>\n

En condicions normals, el parell de prote\u00efnes del metabolisme ossi RANK \/ RANKL actua com a traductor de la informaci\u00f3 proporcionada per hormones sexuals com la progesterona i envia una senyal que estimula el desenvolupament de la gl\u00e0ndula mam\u00e0ria. Aix\u00f2 es d\u00f3na en totes les dones durant l’embar\u00e0s i durant el cicle menstrual, per\u00f2 si la via de senyalitzaci\u00f3 es desregula, les c\u00e8l\u00b7lules mam\u00e0ries comencen a dividir-se i multiplicar-se i no moren quan ho haurien de fer, el que d\u00f3na lloc al c\u00e0ncer de mama. <\/p>\n

La troballa publicada a Cancer Research <\/em>\u00e9s considerada com un gran aven\u00e7 perqu\u00e8 els inhibidors de la via de senyalitzaci\u00f3 RANK ja estan aprovats per a \u00fas cl\u00ednic. De fet, aquest tipus d’inhibidors s’utilitzen actualment per tractar l’osteoporosi i en la prevenci\u00f3 d’esdeveniments relacionats amb l’esquelet en pacients que pateixen de met\u00e0stasis \u00f2ssies a causa dels tumors. “Com aquests experiments precl\u00ednics suggereixen, at\u00e8s que la via de RANKL \/ RANK juga un paper important en el desenvolupament del c\u00e0ncer de mama primari, els inhibidors podrien servir com una diana potencial per a la prevenci\u00f3 i el tractament de c\u00e0ncer de mama”, conclou la investigadora.<\/p>\n","protected":false},"excerpt":{"rendered":"
