Un estudi publicat a la revista cient\u00edfica<\/p>\n

Journal of Investigative Dermatology<\/em><\/a>ha identificat una diana terap\u00e8utica potencial per al tractament de la psoriasi, una malaltia inflamat\u00f2ria cr\u00f2nica de la pell que afecta fins a un 2 % de la poblaci\u00f3 i que encara no t\u00e9 cura. El treball demostra que el gen<\/p>\n

TREX2 t<\/em>\u00e9 un paper rellevant en la resposta inflamat\u00f2ria que es desencadena durant la malaltia. La recerca, liderada per investigadors del Departament de Patologia i Terap\u00e8utica Experimental de la<\/p>\n

contribueix a augmentar el coneixement sobre aquesta malaltia inflamat\u00f2ria complexa i multifactorial i obre la porta a desenvolupar noves estrat\u00e8gies terap\u00e8utiques. En l\u2019estudi, tamb\u00e9 hi participen experts dels<\/p>\n

<\/span>Centres Cient\u00edfics i Tecnol\u00f2gics de la UB (CCiTUB),<\/a><\/p>\n

l\u2019Institut de Medicina Predictiva i Personalitzada del C\u00e0ncer (IMPPC), l\u2019Hospital del Mar, i la Universitat de Wake Forest (Estats Units).<\/span> El gen<\/p>\n

<\/span>TREX2<\/em><\/p>\n

codifica per una exonucleasa que s\u2019havia relacionat amb el manteniment de l\u2019estabilitat gen\u00f2mica, i amb determinats mecanismes tant de reparaci\u00f3 com de degradaci\u00f3 de l\u2019ADN. En estudis anteriors, el grup de recerca va observar que aquesta prote\u00efna s\u2019expressava espec\u00edficament en la pell. A m\u00e9s a m\u00e9s, l\u2019estudi del fenotip de ratolins deficients per aquesta prote\u00efna revela que la p\u00e8rdua de<\/p>\n

<\/span>TREX2<\/p>\n

<\/em>provoca una major susceptibilitat a la carcinog\u00e8nesi de pell indu\u00efda per genot\u00f2xics com ara la radiaci\u00f3 ultraviolada (UV).<\/span> Per aprofundir en el paper de<\/p>\n

<\/em>en l\u2019home\u00f2stasi i patog\u00e8nesi de la pell, els investigadors van voler con\u00e8ixer el paper d\u2019aquesta nucleasa en la psoriasi. Inicialment, van analitzar diferents bi\u00f2psies de pacients i de persones sense la malaltia. Despr\u00e9s, utilitzant diferents models de psoriasi en ratol\u00ed, van comparar l\u2019evoluci\u00f3 de la malaltia en ratolins normals i deficients per<\/p>\n

<\/span>Trex2<\/p>\n

<\/em>per inferir la funci\u00f3 d\u2019aquesta mol\u00e8cula. \u00abEls resultats mostren que<\/p>\n

<\/span>Trex2<\/em>tindria un paper rellevant en la degradaci\u00f3 de l\u2019ADN en l\u2019epidermis psori\u00e0sica, en promoure la resposta inflamat\u00f2ria caracter\u00edstica d\u2019aquesta malaltia\u00bb, explica Concepci\u00f3 Soler, responsable de l\u2019estudi i professora del Departament de Patologia i Terap\u00e8utica Experimental de la Universitat de Barcelona.<\/span> L\u2019estudi mostra que la pell de pacients amb psoriasi experimenta un fort augment de l\u2019expressi\u00f3 de<\/p>\n

<\/span>TREX2<\/em><\/p>\n

que es concentra en el nucli cel\u00b7lular dels queratin\u00f2cits \u2014les c\u00e8l\u00b7lules m\u00e9s abundants de la pell\u2014, que proliferen, moren i es diferencien de manera aberrant. Per tant, els resultats suggereixen un paper important d\u2019aquest gen en el processament de l\u2019ADN. \u00abEn el nostre treball, posem de manifest el paper fonamental de<\/p>\n

<\/span>TREX2<\/em><\/p>\n

en la degradaci\u00f3 de l\u2019ADN i la conseq\u00fcent mort cel\u00b7lular del queratin\u00f2cit, fet que influeix en la resposta immunit\u00e0ria a la pell. L\u2019alliberament de diversos senyals per part de les c\u00e8l\u00b7lules de la pell que s\u2019estan morint contribueix a generar i amplificar la resposta immunit\u00e0ria cr\u00f2nica i la hiperproliferaci\u00f3 i diferenciaci\u00f3 anormal de l\u2019epidermis\u00bb, explica el primer autor del treball, Joan Manils, membre del Departament esmentat i ara investigador postdoctoral a l\u2019Institut de Ci\u00e8ncies Biom\u00e8diques del Trinity College (Irlanda).<\/span> Els resultats situen<\/p>\n

com una potencial diana terap\u00e8utica per abordar aquesta malaltia amb una estrat\u00e8gia diferent i m\u00e9s focalitzada. \u00abLa majoria de tractaments actuals estan dirigits a bloquejar l\u2019acci\u00f3 del sistema immunitari, i si b\u00e9 donen bons resultats, s\u00f3n tractaments de car\u00e0cter cr\u00f2nic i comprometen la resposta immunit\u00e0ria del malalt\u00bb, explica Francisco Ciruela, coautor de l\u2019estudi, professor del Departament de Patologia i Terap\u00e8utica Experimental de la UB i cap del Grup de Recerca de Neurofarmacologia i Dolor a l\u2019IDIBELL.<\/span> El proper objectiu dels investigadors ser\u00e0 desxifrar el mecanisme d\u2019acci\u00f3 complet de<\/p>\n
