Investigadors del Grup de Diferenciaci\u00f3 Cel\u00b7lular dins del Programa d\u2019Epigen\u00e8tica i Biologia del c\u00e0ncer de<\/p>\n

l\u2019Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), liderats per la investigadora Maribel Parra, han identificat un repressor transcripcional, la histona desacetilasa HDAC7, implicat en la generaci\u00f3 i la identitat dels limf\u00f2cits B, les c\u00e8l\u00b7lules encarregades de crear anticossos en el nostre sistema immunitari. Els resultats de l\u2019estudi, publicat a The Journal of Experimental Medicine, demostren que HDAC7 es necess\u00e0ria per la formaci\u00f3 dels limf\u00f2cits B. <\/p>\n

Les c\u00e8l\u00b7lules mare de la sang passen per un sistema molt complex de diferenciaci\u00f3 per formar les diferents c\u00e8l\u00b7lules sangu\u00ednies. Existeixen dues l\u00ednies principals: la limfoide, que acaba donant lloc a les c\u00e8l\u00b7lules T, els limf\u00f2cits B i les c\u00e8l\u00b7lules NK, i la mieloide que formar\u00e0 els eritr\u00f2cits (gl\u00f2buls vermells) o els macr\u00f2fags entre d\u2019altres. Segons ha explicat Maribel Parra, per tal que es donin aquests passos s\u2019han de produir una s\u00e8rie de canvis en el programa g\u00e8nic de les c\u00e8l\u00b7lules. Existeixen factors de transcripci\u00f3 que activen gens de cada llinatge cel\u00b7lular per\u00f2 tamb\u00e9 existeixen repressors transcripcionals que impedeixen que s\u2019expressin gens \u201cinapropiats\u201d, que s\u00f3n caracter\u00edstics d\u2019un altre llinatge. Fins ara, la identitat i mecanismes d\u2019acci\u00f3 d\u2019aquests repressors era molt poc coneguda. El grup de la Dra. Parra estudia precisament el paper d\u2019una fam\u00edlia de prote\u00efnes que s\u00f3n repressores transcripcionals en el sistema hematopoi\u00e8tic. Es tracta de les histones desacetilases (HDACs). <\/p>\n

En aquest treball els investigadors demostren que el paper d\u2019una d\u2019aquestes prote\u00efnes, l\u2019HDAC7, en limf\u00f2cits B \u00e9s reprimir gens \u2018inapropiats\u2019, caracter\u00edstics d\u2019altres tipus cel\u00b7lulars com els macr\u00f2fags i el limf\u00f2cits T. Els investigadors han utilitzat un model de ratol\u00ed deficient en HDAC7 en progenitors de c\u00e8l\u00b7lules B (pro-B). Han observat que en abs\u00e8ncia de HDAC7, els ratolins presenten un bloqueig en la formaci\u00f3 de limf\u00f2cits B. Els investigadors han trobat que HDAC7 s\u2019uneix a gens caracter\u00edstics d\u2019altres tipus cel\u00b7lulars conduint a la seva repressi\u00f3 transcripcional. <\/p>\n

Maribel Parra ha explicat que \u201ca trav\u00e9s d\u2019aquets mecanisme de repressi\u00f3 transcripcional, la c\u00e8l\u00b7lula B s\u2019assegura que no s\u2019expressin gens d\u2019altres tipus cel\u00b7lulars, mantenint la seva identitat limfoide\u201d. Tamb\u00e9 assenyala que \u201cdonada la seva import\u00e0ncia en la biologia dels limf\u00f2cits B, la funci\u00f3 d\u2019HDAC7 podria estar alterada en malalties hematol\u00f2giques. De fet l\u2019any passat vam publicar un article en el que veiem que l\u2019expressi\u00f3 d\u2019HDAC7 estava desregulada en tipus concrets de leuc\u00e8mies i limfomes de c\u00e8l\u00b7lules B.\u201d <\/p>\n
