Investigadors del grup de TGF-beta i C\u00e0ncer de l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), en col\u00b7laboraci\u00f3 amb el King’s College de Londres, han demostrat el paper de la NADPH oxidasa NOX4 com a inhibidor de la transici\u00f3 epiteli-ameboide, un proc\u00e9s que contribueix a la migraci\u00f3 i invasi\u00f3 de les c\u00e8l\u00b7lules tumorals. L’estudi acaba de publicar-se a la revista Oncogene. <\/p>\n
En estudis anteriors, els investigadors ja havien proposat NOX4 com un supressor tumoral en el fetge, ja que inhibeix la proliferaci\u00f3 cel\u00b7lular. “En aquest treball, vam demostrar que NOX4 \u00e9s tamb\u00e9 un important inhibidor de la invasi\u00f3 i met\u00e0stasi de les c\u00e8l\u00b7lules tumorals del fetge”, explica la Dra. Isabel Fabregat, l\u00edder del grup de recerca de l’IDIBELL. <\/p>\n
Els estudis in vitro<\/em> indiquen que el silenciament de NOX4 en c\u00e8l\u00b7lules hep\u00e0tiques tumorals indueix un moviment migratori que es coneix com ameboide. El moviment ameboide, relacionat amb la contractilitat de les c\u00e8l\u00b7lules, est\u00e0 regulat per la fam\u00edlia de prote\u00efnes Rho; una major expressi\u00f3 d’aquestes prote\u00efnes d\u00f3na lloc a aquest tipus de moviment, associat amb met\u00e0stasis agressives. <\/p>\n En l’estudi en pacients, s’observa que un important nombre de casos tenen silenciat NOX4; a m\u00e9s, aquells pacients amb una baixa expressi\u00f3 de NOX4 i una alta expressi\u00f3 de prote\u00efnes Rho presenten molt pitjor pron\u00f2stic. “Aix\u00f2 atorga al nostre estudi in vitro<\/em> una rellev\u00e0ncia translacional, ja que proposa nous biomarcadors pron\u00f2stics per a aquest tipus de c\u00e0ncer”, comenta la investigadora de l’IDIBELL. <\/p>\n NOX4 est\u00e0 regulat a nivell transcripcional per la citocina TGF-beta, estudiada pel grup de recerca des de fa m\u00e9s de 15 anys. Se sap que TGF-beta actua com supressor tumoral en estadis primerencs, per\u00f2 passa a ser un inductor en estadis tardans. D’acord amb estudis previs del grup, quan el nivell d’expressi\u00f3 de NOX4 \u00e9s alt, TGF-beta s’estaria comportant com supressor i la utilitzaci\u00f3 de f\u00e0rmacs inhibidors de TGF-beta seria contraproduent. No obstant, el present estudi suggereix que quan el nivell d’expressi\u00f3 de NOX4 \u00e9s baix i el de prote\u00efnes Rho \u00e9s alt, els pacients podrien ser candidats per a aquest tipus de f\u00e0rmacs. “Aix\u00ed doncs, la baixa expressi\u00f3 de NOX4 no nom\u00e9s podria emprar-se com a marcador de mal pron\u00f2stic, sin\u00f3 tamb\u00e9 com a marcador per a la utilitzaci\u00f3 de f\u00e0rmacs inhibidors de TGF-beta”, conclou la Dra. Fabregat. <\/p>\n El treball, la principal autora del qual \u00e9s Eva Crosas-Molist, ha estat codirigit entre Victoria Sanz Moreno, del King’s College, i Isabel Fabregat per part de l’IDIBELL.
