La fosforilaci\u00f3 d\u2019EPRS condiciona les respostes metab\u00f2liques dependents de S6K1 relacionades amb l’obesitat i l’envelliment<\/strong> Els investigadors de l’Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) i de la Universitat de Barcelona (UB) George Thomas, cap del grup de recerca en Metabolisme i el C\u00e0ncer, i Sara Kozma, publiquen aquesta setmana a la revista Nature<\/em> un article sobre el paper de la quinasa S6K1 en el canvi de funci\u00f3 de la glutamil-prolil-tRNA sintetasa (EPRS) d\u2019inhibidora de la traducci\u00f3 a transportadora d’\u00e0cids grassos. L’estudi es va dur a terme en col\u00b7laboraci\u00f3 amb investigadors de la Cleveland Clinic i la Universitat d’Illinois, i podria conduir potencialment al desenvolupament de biomarcadors i ter\u00e0pies dirigides contra l’obesitat i l’envelliment. <\/p>\n Estudis anteriors dels doctors Thomas i Kozma de l\u2019IDIBELL-UB ja havien demostrat que S6K1 exerceix un paper cr\u00edtic en l’expansi\u00f3 dels teixits grassos mitjan\u00e7ant la regulaci\u00f3 transcripcional de la diferenciaci\u00f3 de les c\u00e8l\u00b7lules mare mesenquimals (MSC) en adip\u00f2cits (c\u00e8l\u00b7lules de greix). “Mentrestant”, explica el Dr. Thomas, “Paul Fox es va posar en contacte amb nosaltres proposant una possible connexi\u00f3 entre S6K1 i l’enzim EPRS. Junts vam descobrir que la fosforilaci\u00f3 <\/p>\n de S6K1 \u00e9s necess\u00e0ria per alliberar EPRS del seu paper habitual en la s\u00edntesi de prote\u00efnes i formar el complex inhibidor de la traducci\u00f3 d’interfer\u00f3 \u0263 (GAIT), que bloqueja la traducci\u00f3 de gens regulats per l\u2019interfer\u00f3”. <\/p>\n “Aquest canvi de funci\u00f3 es determinant, ja que paral\u00b7lelament vam observar que EPRS interv\u00e9 en la localitzaci\u00f3 de la prote\u00efna de transport d’\u00e0cids grassos 1 (FATP1) dins la membrana, estimulant la captaci\u00f3 d’\u00e0cids grassos de cadena llarga”, afegeix la Dra. Kozma. “Aix\u00f2 va confirmar el doble paper de S6K1 en la seva contribuci\u00f3 a l’adipositat i l’envelliment. De fet, els ratolins amb EPRS sense fosforilar presenten les mateixes caracter\u00edstiques f\u00edsiques (fenotip) que els ratolins deficients en S6K1: pes corporal baix, massa de teixit adip\u00f3s redu\u00efda i augment de l’esperan\u00e7a de vida. Aquests resultats reforcen el concepte que S6K1 pot servir com a biomarcador de l’obesitat o com a diana de f\u00e0rmacs contra l’obesitat”. <\/p>\n La Dra.
