Recents resultats del grup de recerca en mort cel\u00b7lular de l’Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), liderats per la Dra. Cristina Mu\u00f1oz-Pinedo, han perm\u00e8s caracteritzar el proc\u00e9s de mort cel\u00b7lular per falta de nutrients, en el qual el reticle endopl\u00e0smic juga un paper destacat. El seu treball, publicat en portada per la revista Molecular and Cellular Biology, s’emmarca en el context de TRAIN-ERs<\/em>, una acci\u00f3 europea col\u00b7laborativa que estudia les malalties associades a aquest org\u00e0nul cel\u00b7lular.<\/p>\n

“Normalment, el proc\u00e9s de mort cel\u00b7lular programada, anomenat apoptosi, segueix una via bioqu\u00edmica relacionada amb la permeabilitzaci\u00f3 de les mitoc\u00f2ndries; per\u00f2, hem observat que en els casos de mort cel\u00b7lular per falta de nutrients (glucosa), les c\u00e8l\u00b7lules es moren d’una forma inesperada, seguint un proc\u00e9s similar al que es derivaria d’una resposta immune”, explica la Dra. Cristina Mu\u00f1oz- Pinedo, darrera autora de l’estudi.<\/p>\n

En tractaments relacionats amb la mort cel\u00b7lular, com la quimioter\u00e0pia, \u00e9s la via mitocondrial la qual s’activa. En canvi, en abs\u00e8ncia de glucosa, les c\u00e8l\u00b7lules activen els anomenats “receptors de mort” en la seva membrana, que normalment fan servir els limf\u00f2cits del sistema immunitari per atacar i destruir c\u00e8l\u00b7lules infectades.<\/p>\n

Els investigadors de l’IDIBELL han aconseguit relacionar l’activaci\u00f3 d’aquests receptors de membrana amb el reticle endopl\u00e0smic, un org\u00e0nul cel\u00b7lular que interv\u00e9 en funcions relacionades amb la s\u00edntesi de prote\u00efnes, el metabolisme de l\u00edpids i alguns esteroides, aix\u00ed com el transport intracel\u00b7lular. “Davant l’estr\u00e8s produ\u00eft per la falta de nutrients, el reticle d\u00f3na el senyal d’alarma que desencadena l’aparici\u00f3 dels receptors de mort en la membrana”, comenta la Dra. Mu\u00f1oz-Pinedo.<\/p>\n

“D’acord amb els nostres resultats in vitro<\/em>, suposem que aix\u00ed \u00e9s com moren les c\u00e8l\u00b7lules tumorals situades al centre d’un tumor, l’anomenat nucli necr\u00f2tic, ja que en aquesta zones mai arriben suficients nutrients”, afegeix la investigadora de l’IDIBELL. “D’altra banda, en les isqu\u00e8mies, a m\u00e9s de per manca d’oxigen tamb\u00e9 es d\u00f3na mort cel\u00b7lular per falta de nutrients; per tant, tamb\u00e9 podr\u00edem relacionar aquest proc\u00e9s a nivell bioqu\u00edmic amb l’activitat del reticle endopl\u00e0smic”.<\/p>\n

El treball ha estat possible gr\u00e0cies al finan\u00e7ament rebut de la xarxa europea Marie Curie TRAIN-ERs<\/em>, que d\u00f3na suport i forma a joves investigadors per desenvolupar estrat\u00e8gies per al tractament de les malalties associades a l’estr\u00e8s del reticle endopl\u00e0smic. “Dins de TRAIN-ERs<\/em>, l’objectiu del nostre grup d’investigaci\u00f3 era estudiar el paper dels senyals d’estr\u00e8s del reticle endopl\u00e0smic davant la falta de nutrients, i hem demostrat que \u00e9s essencial”, comenta Mu\u00f1oz-Pinedo.<\/p>\n

L’estr\u00e8s del reticle endoplasm\u00e0tic (ER) est\u00e0 emergint recentment com una caracter\u00edstica comuna en la patologia de nombroses malalties, entre les que s\u2019inclouen c\u00e0ncer, trastorns neurodegeneratius, s\u00edndromes metab\u00f2lics i malalties inflamat\u00f2ries, afectant anualment a milions de pacients a tot el m\u00f3n i suposant una enorme c\u00e0rrega econ\u00f2mica per el sector sanitari.<\/p>\n","protected":false},"excerpt":{"rendered":"
