Investigadors del programa Oncobell de l’Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) -Institut Catal\u00e0 d’Oncologia (ICO) han descobert un nou rol dels ribosomes 40S lliures com a guardians de la informaci\u00f3 gen\u00e8tica necess\u00e0ria per a sintetitzar-se. Aquest mecanisme, que es basa en un complex foramt per ribosomes 40S lliures i la prote\u00efna d’uni\u00f3 a ARN LARP1, \u00e9s independent del paper dels ribosomes 40S en la s\u00edntesi de prote\u00efnes, i pot ser potencialment dirigit com a ter\u00e0pia contra el c\u00e0ncer. A m\u00e9s a m\u00e9s, conjuntament amb investigadors de l’Institut d’Investigacions August Pi i Sunyer (IDIBAPS), l\u2019equip de recerca tamb\u00e9 ha descobert que el complex 40S-LARP1 pot ser un punt potencial d’intervenci\u00f3 per al tractament de la s\u00edndrome 5q-, una malaltia gen\u00e8tica espor\u00e0dica rara, causalment vinculada a la p\u00e8rdua de LARP1 i ribosomes 40S lliures. <\/p>\n

Els ribosomes s\u00f3n les f\u00e0briques de prote\u00efnes de la c\u00e8l\u00b7lula; s\u00f3n responsables de traduir la informaci\u00f3 gen\u00e8tica inscrita pels ARN missatgers (mARNs) en la seq\u00fc\u00e8ncia d’amino\u00e0cids de les prote\u00efnes. “Hem identificat un mecanisme pel qual un subconjunt de ribosomes 40S pot conservar la informaci\u00f3 gen\u00e8tica en forma d’ARNm necess\u00e0ria per produir r\u00e0pidament nous ribosomes i els factors accessoris necessaris en el proc\u00e9s de la traducci\u00f3”, explica el Dr. Antoni Gentilella, primer autor de l’estudi i professor de la Universitat de Barcelona. “En protegir aquesta informaci\u00f3, el ribosoma assegura el seu potencial per reproduir-se i proporcionar la capacitat anab\u00f2lica requerida a demanda”. <\/p>\n

“Nom\u00e9s una porci\u00f3 molt petita dels ribosomes 40S est\u00e0 dedicada a aquesta tasca protectora”, afegeix el Dr. George Thomas, l\u00edder del Laboratori de Metabolisme i C\u00e0ncer de l\u2019IDIBELL-ICO i tamb\u00e9 professor de la Universitat de Barcelona. “S’estableixen d’una manera que els permet reproduir-se cada vegada que la c\u00e8l\u00b7lula envia un senyal – oncog\u00e8nica, o mitog\u00e8nica – que els impulsa per fer-ho. Tenint en compte que les c\u00e8l\u00b7lules canceroses han d’augmentar dr\u00e0sticament la quantitat de maquin\u00e0ria ribosomal per proliferar, aquest nou dip\u00f2sit de ribosomes es torna extremadament important.” <\/p>\n

Totes les prote\u00efnes de la c\u00e8l\u00b7lula s\u00f3n sintetitzades pels ribosomes. Aix\u00f2 els fa centrals no nom\u00e9s en el context de la fisiologia cel\u00b7lular normal, sin\u00f3 tamb\u00e9 en el desenvolupament del c\u00e0ncer. Les c\u00e8l\u00b7lules canceroses necessiten cr\u00e9ixer i generar biomassa constantment, de manera que exploten aquest mecanisme per fer-ho; \u00e9s un segell distintiu de la malaltia. Per tant, la inhibici\u00f3 de la biog\u00e8nesi ribos\u00f2mica est\u00e0 actualment sent considerada com a estrat\u00e8gia terap\u00e8utica contra molts c\u00e0ncers. <\/p>\n

Els investigadors de l’IDIBELL han identificat la prote\u00efna LARP1 com a part del complex que s’uneix als mRNAs que codifiquen les prote\u00efnes ribos\u00f2miques protegides pel ribosoma 40S. “Ara tenim una comprensi\u00f3 m\u00e9s profunda d’aquest proc\u00e9s, que pot servir com un important tal\u00f3 d’Aquil\u00b7les a explotar en el tractament del c\u00e0ncer”, assenyala el Dr. Gentilella; “A m\u00e9s de les ter\u00e0pies actuals, atacar el complex del reservori 40S-LARP1 idealment eliminaria aquest grup de mRNAs responsables de generar nous ribosomes”. “Tenim f\u00e0rmacs capa\u00e7os de bloquejar la biog\u00e8nesi ribos\u00f2mica, per\u00f2 molts ARNm anab\u00f2lics encara es conserven, protegits pel complex 40S-LARP1. Per aix\u00f2, quan es deixa el tractament poden ser r\u00e0pidament utilitzats per generar nous ribosomes, reiniciant el c\u00e0ncer. Aix\u00ed doncs, atacar aquest complex resulta un enfocament emocionant, ja que si destru\u00efm el reservori de mARNs, evitarem la biog\u00e8nesi del ribosoma de manera molt m\u00e9s efectiva “, conclou el Dr. Thomas.<\/p>\n","protected":false},"excerpt":{"rendered":"
