El cisplat\u00ed \u00e9s un dels agents quimioterap\u00e8utics m\u00e9s utilitzats pels onc\u00f2legs per al tractament de tumors. La quimioter\u00e0pia basada en cisplat\u00ed i els seus an\u00e0legs \u00e9s molt eficient, tot i que freq\u00fcentment les c\u00e8l\u00b7lules tumorals reajusten els seus mecanismes moleculars i es tornen resistents a la toxicitat del cisplat\u00ed.\u00a0<\/p>\n

El grup de recerca \u201cModels de malaltia en C. elegans<\/em>\u201d de l\u2019IDIBELL, liderat pel Dr. Juli\u00e1n Cer\u00f3n, porta anys utilitzant un cuc mil\u00b7lim\u00e8tric per investigar els reajustaments moleculars que provoquen l\u2019adquisici\u00f3 de resist\u00e8ncia al cisplat\u00ed. En col\u00b7laboraci\u00f3 amb el grup del Dr. Alberto Villanueva (IDIBELL – Institut Catal\u00e0 d\u2019Oncologia (ICO)), que estudia aquests mecanismes en tumors humans implantats en ratolins, i gr\u00e0cies a la tesi doctoral de Francisco Javier Garc\u00eda Rodr\u00edguez, han establert un m\u00e8tode per identificar gens i vies moleculars que intervenen en la resist\u00e8ncia cel\u00b7lular al cisplat\u00ed.<\/p>\n

L\u2019acci\u00f3 t\u00f2xica del cisplat\u00ed \u00e9s molt similar en les c\u00e8l\u00b7lules de tots els animals i, per tant, tamb\u00e9 ho \u00e9s la resposta d\u2019aquestes c\u00e8l\u00b7lules per protegir-se del cisplat\u00ed. El treball de Garc\u00eda Rodr\u00edguez i col\u00b7laboradors obre la possibilitat de regular la resposta cel\u00b7lular al cisplat\u00ed modificant l\u2019activitat de rutes metab\u00f2liques. En altres paraules, \u00e9s un punt de partida per a investigar com la dieta o f\u00e0rmacs comuns que influeixin en el metabolisme de sucres i greixos poden determinar la resposta dels pacients a la quimioter\u00e0pia basada en el cisplat\u00ed.<\/p>\n

Un dels descobriments m\u00e9s impactants d\u2019aquest treball \u00e9s la troballa de la influ\u00e8ncia que t\u00e9 un sol amino\u00e0cid en la resposta d\u2019un organisme multicel\u00b7lular al cisplat\u00ed. Els investigadors de l\u2019IDIBELL van utilitzar la t\u00e8cnica de CRISPR per canviar la identitat d\u2019un sol nucle\u00f2tid del genoma de C. elegans<\/em>, i modificar aix\u00ed la identitat d\u2019un amino\u00e0cid de la prote\u00efna TRXR-1, que \u00e9s hom\u00f2loga a la prote\u00efna humana tiorredoxina reductasa 1 (TRXR1), fent aix\u00ed que l\u2019animal fos resistent a la toxicitat del cisplat\u00ed. Aquest descobriment obre la porta a la validaci\u00f3 en C. elegans<\/em> de SNPs (canvis puntuals en el genoma) humans per valorar de manera personalitzada la influ\u00e8ncia que canvis m\u00ednims en el genoma puguin tenir en la resposta de pacients a la quimioter\u00e0pia.\u00a0<\/p>\n

La publicaci\u00f3 de dos articles liderats per Juli\u00e1n Cer\u00f3n i Alberto Villanueva en aquest mes de juny en les revistes Disease Models and Mechanisms<\/em> i Clinical Cancer Research<\/em> consolida a C. elegans<\/em> com a animal model en l\u2019estudi de mecanismes de resist\u00e8ncia a determinats agents quimioterap\u00e8utics.
