Con\u00e8ixer en detall la biologia d\u2019una prote\u00efna per entendre malalties d\u2019ampli abast: aquest \u00e9s l\u2019abordatge que es va proposar seguir l\u2019equip de recerca de l\u2019IDIBELL (Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge) que lidera la Dra. Mariona Graupera, referent, a nivell mundial, en el camp de la investigaci\u00f3 en formaci\u00f3 de vasos sanguinis (angiog\u00e8nesi) i en malformacions vasculars, que ha comptat amb la participaci\u00f3 d\u2019investigadors de l\u2019Institut Catal\u00e0 d\u2019Oncologia (ICO-L\u2019Hospitalet). Publicat a la revista Nature Communications \u2011d\u2019elevada repercussi\u00f3 i de car\u00e0cter multidisciplinar\u2011, el darrer treball de Graupera, impulsat per la Fundaci\u00f3 Banc\u00e0ria \u201cla Caixa\u201d, posa de relleu la import\u00e0ncia de la recerca b\u00e0sica i mecan\u00edstica a l\u2019hora d\u2019entendre el paper deteminant d\u2019un gen i d\u2019una prote\u00efna en un context patol\u00f2gic, un estadi del tot imprescindible si volem realitzar amb efectivitat i \u00e8xit la translaci\u00f3 del coneixement cient\u00edfic vers una pr\u00e0ctica cl\u00ednica que contribueixi a la millora de la salut de les persones.<\/p>\n

Nom\u00e9s en el cas de les malformacions venoses, es coneix que una de cada 5.000 persones neixen amb aquesta patologia, i no existeix, de moment, cap cura totalment efectiva. Aquestes malformacions es caracteritzen per la pres\u00e8ncia de vasos sanguinis dilatats i deformes, que tant poden apar\u00e8ixer sota la pell, causant dolor i desfiguraci\u00f3, com profunds en el cos, donant lloc a diversos problemes greus de salut.<\/p>\n

Estudis previs del grup de Graupera, entre d\u2019altres, ja demostraren que un seguit de mutacions en el gen PIK3CA, les que provoquen un guany de funci\u00f3 (o una sobreactivaci\u00f3) de la prote\u00efna per a la qual codifica (i que rep el nom de PI3K\uf061), estan implicades tant en c\u00e0ncer com en patologies de sobrecreixement, com ara el creixement desmesurat dels vasos sanguinis i les malformacions venoses a les quals ens refer\u00edem.<\/p>\n

Per entendre el mecanisme a trav\u00e9s del qual aquests errors gen\u00e8tics desemboquen en la manifestaci\u00f3 d\u2019aquestes patologies severes, l\u2019estudi recentment publicat s\u2019ha proposat descriure la biologia completa de PIK3CA.<\/p>\n

Tot treballant amb tres models experimentals que han aportat informaci\u00f3 complement\u00e0ria, els investigadors han elaborat un mapa de les funcions d\u2019aquest gen, i han descrit els efectors biol\u00f2gics mitjan\u00e7ant els quals les realitza. En concret, s\u2019han utilitzat ratolins, peixos zebra i c\u00e8l\u00b7lules en cultiu, i en tots tres models s\u2019ha abordat el problema provocant una p\u00e8rdua de funci\u00f3 del gen d\u2019inter\u00e8s, amb la finalitat de descriure els processos per als quals resulta imprescindible la seva pres\u00e8ncia, i que condueixen a la formaci\u00f3 dels vasos sanguinis.<\/p>\n

Els darrers resultats obtinguts se sumen al coneixement que aportaren estudis anteriors, publicats en els darrers anys. En un d\u2019aquests, els investigadors modificaren ratolins gen\u00e8ticament per introduir-los una mutaci\u00f3 de guany de funci\u00f3 de PIK3CA, que result\u00e0 en el desenvolupament, de forma inesperada, de malformacions venoses. Segons Graupera, \u201ccaracteritzar, amb el m\u00e0xim detall possible, els mecanismes d\u2019acci\u00f3 d\u2019aquest gen ens apropa a l\u2019estrat\u00e8gia terap\u00e8utica dels malalts que presenten mutacions activadores de PIK3CA i un creixement excessiu dels vasos sanguinis\u201d.
