Els tumors, per cr\u00e9ixer i proliferar, realitzen una gran despesa energ\u00e8tica. Les c\u00e8l\u00b7lules canceroses s\u00f3n, de fet, addictes al principal combustible del metabolisme cel\u00b7lular, la glucosa. La maquin\u00e0ria molecular que degrada aquest sucre per obtenir energia (els anomenats enzims glucol\u00edtics) est\u00e0 sobreexpressada en c\u00e8l\u00b7lules tumorals. Per\u00f2 aquesta no \u00e9s l\u2019\u00fanica caracter\u00edstica que diferencia el metabolisme de les c\u00e8l\u00b7lules tumorals del de les normals. En efecte, la tumorig\u00e8nesi implica grans canvis metab\u00f2lics a les c\u00e8l\u00b7lules, englobats sota el concepte de reprogramaci\u00f3 metab\u00f2lica.<\/p>\n

L’equip de recerca de l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) i del Campus de Bellvitge de la Universitat de Barcelona (UB) ha donat un important pas endavant en el coneixement del mecanisme molecular que reprograma la c\u00e8l\u00b7lula en el proc\u00e9s de tumorig\u00e8nesi. El treball, publicat a la revista iScience, desxifra la via a trav\u00e9s de la qual la glucosa potencia el creixement cel\u00b7lular.<\/p>\n

Per poder dividir-se, les c\u00e8l\u00b7lules han de cr\u00e9ixer i assolir una mida cr\u00edtica. Promovent processos biosint\u00e8tics -com la producci\u00f3 de prote\u00efnes i l\u00edpids-, duplicant el material gen\u00e8tic i suprimint vies de degradaci\u00f3 de les biomol\u00e8cules, les c\u00e8l\u00b7lules es preparen per proliferar. Aquests processos s\u00f3n regulats per una prote\u00efna clau: l’anomenada mTOR, que exerceix un paper nodal en la reprogramaci\u00f3 metab\u00f2lica.<\/p>\n

L\u2019equip de recerca, liderat pel Dr. Albert Tauler, membre tamb\u00e9 de la Facultat de Farm\u00e0cia i Ci\u00e8ncies de la Salut de la UB, ha demostrat de quina forma s’acobla, molecularment, la disponibilitat dels nutrients que han de permetre el creixement cel\u00b7lular, i les senyals gen\u00e8tiques, com \u00e9s el cas de l’oncogen E2F1, que promouen la divisi\u00f3 descontrolada de la c\u00e8l\u00b7lula tumoral. Un mecanisme que mant\u00e9 mTOR al centre de la reprogramaci\u00f3.<\/p>\n

En concret, el treball descriu la interacci\u00f3 activadora de dos enzims glucol\u00edtics (la PFKFB3 i la PFK1) amb el complex mTOR, que conflueixen a la superf\u00edcie d’uns org\u00e0nuls cel\u00b7lulars anomenats lisosomes. Els lisosomes s\u00f3n org\u00e0nuls recoberts per una membrana, que contenen una gran quantitat d’enzims capa\u00e7os de degradar les macromol\u00e8cules. La translocacio d\u2019mTOR a la superf\u00edcie dels lisosomes, imprescindible per a la seva activaci\u00f3 i funci\u00f3, est\u00e0 activada per l\u2019activitat d\u2019aquest dos enzims glucol\u00edtics, un cop han estat reclutats, tamb\u00e9, a la membrana dels lisosomes.<\/p>\n

Els resultats recentment publicats demostren, doncs, que la PFKFB3 i la PFK1 s\u00f3n el nexe entre la disponibilitat de glucosa i l’activaci\u00f3 d’mTOR. Alhora, evidencien la import\u00e0ncia dels lisosomes com a plataformes de senyalitzaci\u00f3 essencials per la regulaci\u00f3 de la reprogramaci\u00f3 metab\u00f2lica, inherent a la tumorig\u00e8nesi.<\/p>\n

mTOR ja \u00e9s, actualment, una diana terap\u00e8utica per combatre el c\u00e0ncer. La rellev\u00e0ncia de l’estudi publicat rau en la descripci\u00f3 d’una nova via de regulaci\u00f3 d’aquesta prote\u00efna que permetr\u00e0 explorar nous abordatges terap\u00e8utics d’una de les malalties amb m\u00e9s impacte en la societat actual.<\/p>\n","protected":false},"excerpt":{"rendered":"
