L\u2019adrenoleucodistr\u00f2fia lligada al cromosoma X (X ALD) \u00e9s una malaltia gen\u00e8tica minorit\u00e0ria en qu\u00e8 hi ha una acumulaci\u00f3 d\u2019\u00e0cids grassos de cadena molt llarga a la sang i al teixit nervi\u00f3s, i on la mielina de les neurones es fa malb\u00e9. Les persones que la pateixen (1 de cada 14.700 nounats) poden tenir, entre d\u2019altres, problemes cerebrals i de mobilitat, a m\u00e9s d\u2019alteracions hormonals. La causa \u00e9s una defici\u00e8ncia en el gen ABCD1, que codifica la prote\u00efna de la adrenoleucodistr\u00f2fia (ALDP), que transporta els \u00e0cids grassos de cadena molt llarga cap a l\u2019interior dels peroxisomes. Aquests org\u00e0nuls tenen un paper molt important en la degradaci\u00f3 dels l\u00edpids i el seu posterior aprofitament cel\u00b7lular.<\/p>\n

En aquest treball, s\u2019ha identificat i caracteritzat en el cuc Caenorhabditis elegans (C. elegans) un model de la malaltia, al qual li manca la prote\u00efna an\u00e0loga (hom\u00f2loga) a l\u2019ALDP humana. L\u2019equip de recerca, co-liderat per les Dres Aurora Pujol i Esther Dalf\u00f3, ha analitzat a nivell cel\u00b7lular les conseq\u00fc\u00e8ncies d\u2019aquest d\u00e8ficit i ha comprovat que, tal com passa en l\u2019hum\u00e0 i en els models existents de ratol\u00ed, hi ha una acumulaci\u00f3 d\u2019\u00e0cids grassos de cadena molt llarga, canvis en el metabolisme lip\u00eddic, desequilibris oxidatius als mitocondris i alteracions neuronals. El model permetr\u00e0 avan\u00e7ar m\u00e9s r\u00e0pidament en l\u2019estudi d\u2019aquesta malaltia, per a la qual actualment no hi ha tractament.<\/p>\n

En el treball, publicat a Free Radical Biology and Medicine, hi han participat investigadors d\u2019ICREA, de l\u2019Institut d\u2019Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), el Centro de Investigaci\u00f3n Biom\u00e9dica en red (CIBERER), de l\u2019Institut de Neuroci\u00e8ncies de la Universitat Aut\u00f2noma de Barcelona (INc-UAB), de la Facultat de Medicina de la Universitat de Vic – Universitat Central de Catalunya (UVic-UCC), el Kennedy Krieger Institute, de Baltimore, el MRC Mitochondrial Biology Unit, de Cambridge, i el Instituto de Biomedicina de Sevilla (IBIS).<\/p>\n

Una eina gen\u00e8tica molt valuosa<\/h4>\n

\u201cAquest model d\u2019adrenoleucodistr\u00f2fia en C. elegans \u00e9s una eina gen\u00e8tica molt valuosa que ens permetr\u00e0 estudiar els mecanismes implicats en la malaltia i trobar dianes farmacol\u00f2giques m\u00e9s \u00e0gilment que amb altres models animals, com els ratolins, que s\u00f3n molt m\u00e9s complexos i impliquen un proc\u00e9s m\u00e9s cost\u00f3s, tant en termes de temps com econ\u00f2mics\u201d, explica Esther Dalf\u00f3, que ara lidera el grup de C elegans models of diseases entre \u00a0l\u2019INc-UAB i la Facultat de Medicina de la UVic-UCC.<\/p>\n

De fet, l\u2019equip de recerca ha comen\u00e7at ja a obtenir els primers resultats, que, tot i que molt preliminars, apunten a que serien les c\u00e8l\u00b7lules glials, que es troben al cervell junt amb les neurones, les primeres responsables de produir les alteracions neurol\u00f2giques associades a la malaltia.<\/p>\n

\u201cAquest nou model animal ens ha servit per confirmar que l\u2019estr\u00e8s oxidatiu originat als mitocondris (els org\u00e0nuls productors d\u2019energia de les c\u00e8l\u00b7lules) \u00e9s el principal causant del dany neuronal en l\u2019adrenoleucodistr\u00f2fia, i aquest mecanisme de dany est\u00e0 conservat des de el cuc al pacient. Les dades apunten noves vies terap\u00e8utiques, com els antioxidants mitocondrials, per tractar una malaltia del peroxisoma\u201d, explica Aurora Pujol, professora ICREA a l\u2019IDIBELL.<\/p>\n

C. elegans, neurones i gens com els humans<\/h4>\n

El cuc de terra C. elegans \u00e9s \u00e0mpliament utilitzat en la recerca biom\u00e8dica perqu\u00e8, malgrat ser molt senzill i petit (mesura nom\u00e9s 1 mm), t\u00e9 la majoria d’estructures dels animals m\u00e9s complexos, i compateix gran part de les vies metab\u00f2liques amb nosaltres. A m\u00e9s, un 40% del seu genoma compr\u00e8n gens hom\u00f2legs als nostres, i t\u00e9 representaci\u00f3 de totes les poblacions neuronals presents al cervell hum\u00e0.<\/p>\n

La manipulaci\u00f3 gen\u00e8tica d\u2019aquest organisme per crear models transg\u00e8nics que permetin estudiar malalties, com l\u2019X ALD, \u00e9s m\u00e9s r\u00e0pida que amb altres models animals com podrien ser les rates o els ratolins. Tot plegat, junt amb trets com la transpar\u00e8ncia del seu cos, una reproducci\u00f3 nombrosa -fins a 300 descendents- i un baix cost, el fan una eina ideal per utilitzar als laboratoris.<\/p>\n","protected":false},"excerpt":{"rendered":"
