El Dr Alberto Ortega, investigador principal del grup de recerca en trastorns del neurodesenvolupament de l\u2019IDIBELL, ha obtingut finan\u00e7ament per participar en un projecte dels Instituts Nacionals de Salut<\/a> (NIH, per les seves sigles en angl\u00e8s) dels Estats Units amb l\u2019objectiu d\u2019investigar com canvia la matriu extracel\u00b7lular de les neurones en diferents etapes del desenvolupament i aix\u00ed avan\u00e7ar en la comprensi\u00f3 de malalties neurodegeneratives. El projecte, que porta per nom “Refinant els sistemes model de medul\u00b7la espinal basats en c\u00e8l\u00b7lules mare pluripotents indu\u00efdes (iPSC) mitjan\u00e7ant la fabricaci\u00f3 de senyals de matriu extracel\u00b7lular programades segons el desenvolupament”, ser\u00e0 coordinat pel professor Evangelos Kiskinis de la Northwestern University a Chicago i tamb\u00e9 comptar\u00e0 amb la participaci\u00f3 de l\u2019IBEC. Durar\u00e0 cinc anys i compta amb un pressupost de m\u00e9s de 2,7 milions de d\u00f2lars, dels quals l’IDIBELL rebr\u00e0 600.000.<\/p>\n

Les tecnologies de reprogramaci\u00f3 cel\u00b7lular i les c\u00e8l\u00b7lules mare pluripotents indu\u00efdes (iPSC) han revolucionat l’estudi del sistema nervi\u00f3s central hum\u00e0, fet que permet crear models per investigar el desenvolupament neuronal i les malalties neurol\u00f2giques. No obstant aix\u00f2, cultivar neurones derivades d’iPSC in vitro<\/em> continua sent un desafiament, ja que no assoleixen la maduresa necess\u00e0ria per estudiar patologies neurodegeneratives. Aix\u00f2 es deu, en part, a la falta d’una matriu extracel\u00b7lular (ECM) adequada, que \u00e9s l’estructura que suporta i regula les c\u00e8l\u00b7lules al cos.<\/p>\n

El \u00a0projecte en q\u00fcesti\u00f3 proposa superar aquest repte investigant com canvia l’ECM en diferents etapes de desenvolupament, des de la infantesa fins a la vellesa, utilitzant la medul\u00b7la espinal de ratolins i humans. A partir d’aquestes troballes, els investigadors dissenyaran matrius artificials que imitin l’ECM natural per millorar la maduraci\u00f3 i l’envelliment de les neurones al laboratori. Aquests aven\u00e7os permetran desenvolupar millors models de medul\u00b7les espinal basats en iPSC, amb aplicacions en l’estudi de malalties i en el desenvolupament de ter\u00e0pies.<\/p>\n

A l\u2019IDIBELL, l\u2019equip del Dr Ortega ser\u00e0 l\u2019encarregat de dur a terme an\u00e0lisis d\u2019espectrometria de masses per determinar canvis en les prote\u00efnes de l\u2019ECM de la medul\u00b7la espinal, un proc\u00e9s molt complex a nivell t\u00e8cnic i que requereix de protocols molt espec\u00edfics. El Dr Ortega ja ha identificat alguns canvis relacionats amb l\u2019edat en l\u2019ECM medul\u00b7lar de ratolins i el seu equip aportar\u00e0 dades de mostres humanes obtingudes gr\u00e0cies a la col\u00b7laboraci\u00f3 amb m\u00faltiples biobancs, entre ells el Biobanc HUB-ICO-IDIBELL. A m\u00e9s, tamb\u00e9 estudiaran l\u2019expressi\u00f3 gen\u00e8tica de les mateixes mostres en col\u00b7laboraci\u00f3 amb el CNAG. Per acabar, l\u2019equip de l\u2019IDIBELL testar\u00e0 l\u2019efecte en la superviv\u00e8ncia de les matrius d\u2019ECM dissenyades per l\u2019IBEC en models 2D i 3D basats en iPSCs. Aquelles que passin aquest test inicial, s\u2019enviaran a la Northwestern University perqu\u00e8 l\u2019equip del Dr Kiskinis en faci la caracteritzaci\u00f3 gen\u00e8tica, bioqu\u00edmica i electrofisiol\u00f2gica necess\u00e0ria.<\/p>\n

El projecte ha estat finan\u00e7at sota el programa R01 dels NIH, un dels majors finan\u00e7adors p\u00fablics de recerca biom\u00e8dica al m\u00f3n, que dona suport a investigacions independents d’alt impacte en \u00e0rees espec\u00edfiques d’inter\u00e8s cient\u00edfic. Aquest tipus de finan\u00e7ament permet a investigadors que es troben en etapes primerenques de la seva carrera investigadora explorar solucions innovadores per abordar desafiaments en la biomedicina.<\/p>\n

La investigaci\u00f3 esmentada en aquesta publicaci\u00f3 ha estat recolzada per l’Institut Nacional sobre l’Envelliment dels Instituts Nacionals de Salut (National Institutes of Health – NIH) sota el n\u00famero de concessi\u00f3 R01AG086270.<\/p>\n
