L’acumulaci\u00f3 de ferro i l’envelliment cel\u00b7lular s’han relacionat amb la fibrosi hep\u00e0tica, una condici\u00f3 en qu\u00e8 el fetge desenvolupa teixit fibr\u00f3s (a mode de cicatriu) a causa d’un dany cr\u00f2nic, com passa a l’hepatitis cr\u00f2nica i el c\u00e0ncer de fetge. Tot i que s’ha identificat la connexi\u00f3 entre aquests dos factors, no s’havia investigat a fons com podrien estar mecan\u00edsticament relacionats. En aquest sentit, un nou estudi, liderat per la investigadora de l’IDIBELL Isabel Fabregat, aborda aquesta q\u00fcesti\u00f3 i explora si un quelant de ferro \u2014\u00e9s a dir, un tipus de f\u00e0rmac que neutralitza el ferro\u2014 podria prevenir la fibrosi hep\u00e0tica en reduir l’envelliment de les c\u00e8l\u00b7lules hep\u00e0tiques.<\/p>\n

Publicat a la revista Cell Death and Disease<\/em>, l’estudi utilitza un model experimental de dany hep\u00e0tic cr\u00f2nic en ratolins, indu\u00eft amb tetraclorur de carboni (CCl4), per examinar com l’acumulaci\u00f3 de ferro es relaciona amb la fibrosi hep\u00e0tica i l’envelliment cel\u00b7lular. Els investigadors han descobert que durant la progressi\u00f3 de la fibrosi hep\u00e0tica, el ferro s’acumula al fetge, cosa que coincideix amb l’aparici\u00f3 d’\u00e0rees fibroses i c\u00e8l\u00b7lules hep\u00e0tiques envellides. Els resultats sorgeixen d’un projecte Retos-Colaboraci\u00f3n finan\u00e7at per l’Ag\u00e8ncia Estatal de Recerca dut a terme per investigadors de l’IDIBELL (Josep Amengual i Isabel Fabregat), del CIC bioGUNE (M Luz Mart\u00ednez-Chantar) \u2014tots dos pertanyents al CIBEREHD\u2014, i de l\u2019IRB Barcelona (Manuel Serrano i Mate Maus, actual l\u00edder del Grup d’Envelliment i C\u00e0ncer de la Vall d’Hebron Institut d’Oncologia \u2013 VHIO).<\/p>\n

Els hepat\u00f2cits, les c\u00e8l\u00b7lules principals del fetge, dels animals tractats van mostrar un perfil d’expressi\u00f3 de gens (conegut com a signatura transcript\u00f2mica) relacionat amb l’acumulaci\u00f3 de ferro i l’envelliment cel\u00b7lular. Aquesta signatura inclo\u00efa tamb\u00e9 l’activaci\u00f3 de gens relacionats amb l’estr\u00e8s oxidatiu, l’activaci\u00f3 la via del TGF-beta i la inhibici\u00f3 del metabolisme oxidatiu.<\/p>\n

El tractament amb deferiprona, un quelant de ferro, va reduir l’acumulaci\u00f3 de ferro, la fibrosi i l’envelliment cel\u00b7lular al fetge dels ratolins. A m\u00e9s, el perfil de les prote\u00efnes secretades per les c\u00e8l\u00b7lules envellides va canviar cap a un perfil m\u00e9s antiinflamatori.<\/p>\n

En experiments amb c\u00e8l\u00b7lules hep\u00e0tiques humanes, es va observar que el ferro s’acumulava en resposta a un agent que indueix envelliment cel\u00b7lular, i que la deferiprona podia prevenir aquest envelliment i els seus efectes associats. Tot i aix\u00f2, la deferiprona no va tenir un efecte significatiu en les c\u00e8l\u00b7lules estrellades hep\u00e0tiques, que tamb\u00e9 juguen un paper en la fibrosi hep\u00e0tica.<\/p>\n

En conclusi\u00f3, l\u2019estudi demostra que l\u2019acumulaci\u00f3 de ferro \u00e9s un factor important en la progressi\u00f3 de la fibrosi hep\u00e0tica i l\u2019envelliment cel\u00b7lular. Les troballes suggereixen que l’\u00fas de quelants de ferro podria ser una estrat\u00e8gia efectiva per alentir o prevenir la fibrosi hep\u00e0tica en reduir l’envelliment cel\u00b7lular als hepat\u00f2cits. Aquest fet podria obrir noves oportunitats per al tractament de malalties hep\u00e0tiques cr\u00f2niques i millorar la qualitat de vida dels pacients afectats.<\/p>\n
