La seva funci\u00f3 endocrina \u00e9s la de secretar al torrent sanguini diverses hormones, com la insulina i el glucag\u00f3, que regulen els nivells de glucosa a la sang. I com a exocrina, secreta al duod\u00e8, a trav\u00e9s del seu conducte, el suc pancre\u00e0tic, que cont\u00e9 bicarbonat i enzims digestius.<\/p>\n
L’equip de la Dra. Rovira ha aconseguit identificar 15 noves poblacions de c\u00e8l\u00b7lules al p\u00e0ncrees, cadascuna amb els seus comportaments i caracter\u00edstiques particulars. Segons declara la investigadora, \u201caquests resultats s\u00f3n molt rellevants: poden ser un gran punt de partida per comprendre millor com es desenvolupen les malalties del p\u00e0ncrees, com per exemple el c\u00e0ncer de p\u00e0ncrees. Per\u00f2, a m\u00e9s, hem observat que les c\u00e8l\u00b7lules dels conductes tenen una gran plasticitat en cultiu i es podrien fer servir com a font de noves c\u00e8l\u00b7lules productores d’insulina en cultius d’organoides per a ter\u00e0pies de medicina regenerativa en diabetis\u201d. Segons la investigadora, les dades recentment publicades senten les bases per a estudis futurs que investiguin el paper de noves poblacions cel\u00b7lulars dels conductes del p\u00e0ncrees en la regeneraci\u00f3 de l’\u00f2rgan i la patog\u00e8nesi de la seva funci\u00f3 exocrina. \u201cSi sabem com es desenvolupa una malaltia\u201d, afegeix Rovira, \u201cpodem dissenyar millors ter\u00e0pies contra ella, m\u00e9s precises i personalitzades\u201d.<\/p>\n

An\u00e0lisi c\u00e8l\u00b7lula a c\u00e8l\u00b7lula<\/strong><\/p>\nEls resultats de l’estudi s’han obtingut mitjan\u00e7ant cultius d’organoides (o mini-\u00f2rgans tridimensionals), models de lesions exocrines i mostres de c\u00e0ncer de p\u00e0ncrees. Per examinar l’expressi\u00f3 g\u00e8nica de c\u00e8l\u00b7lules individuals s’ha fet servir la t\u00e8cnica scRNA-seq (de l’angl\u00e8s, single-cell transcriptomics<\/em>), que \u00e9s capa\u00e7 de mesurar simult\u00e0niament la concentraci\u00f3 d’ARN de milers de gens a cada c\u00e8l\u00b7lula. Gr\u00e0cies a aquests enfocaments experimentals, s’ha pogut caracteritzar exhaustivament l’heterogene\u00eftat ductal del ratol\u00ed en la resoluci\u00f3 unicel\u00b7lular, des de les c\u00e8l\u00b7lules centroacinars fins al conducte principal del p\u00e0ncrees. Tot i aix\u00ed, els investigadors aclareixen que es necessiten nous models animals per investigar, in vivo<\/em> i m\u00e9s a fons, els resultats obtinguts.<\/p>\nLa translaci\u00f3 d’aquestes troballes a l’entorn hum\u00e0 permetria, per una banda, identificar diferents c\u00e8l\u00b7lules dels conductes del p\u00e0ncrees involucrades en l’origen del c\u00e0ncer de p\u00e0ncrees i, en conseq\u00fc\u00e8ncia, avan\u00e7ar cap a una nova estratificaci\u00f3 dels pacients basada en marcadors, i identificar nous objectius terap\u00e8utics. D’altra banda, els resultats obtinguts en cultius d’organoides podrien donar lloc a ter\u00e0pies futures de reempla\u00e7ament cel\u00b7lular per al tractament de la diabetis.<\/p>\n
