El c\u00e0ncer de c\u00f2lon \u00e9s conegut per la seva complexitat i, sobretot, per la seva capacitat per a resistir els tractaments convencionals, per\u00f2 un estudi publicat a la revista Cancer Discovery<\/a>, ha identificat i validat, a nivell precl\u00ednic, una estrat\u00e8gia innovadora que permetria aprofitar l\u2019activaci\u00f3 de les vies oncog\u00e8niques per induir la mort de les c\u00e8l\u00b7lules cancer\u00edgenes.<\/p>\n

Aquest estudi, que ha estat encap\u00e7alat i liderat per l\u2019investigador Rene Bernards de l\u2019institut de recerca NKI (Netherlands Cancer Institute) d\u2019Amsterdam, ha comptat amb la col\u00b7laboraci\u00f3 d\u2019investigadors de l\u2019Institut Catal\u00e0 d\u2019Oncologia (ICO), l\u2019IDIBELL i l\u2019Hospital de Bellvitge.<\/p>\n

Inhibir la prote\u00efna PP2A per a desencadenar la mort de les c\u00e8l\u00b7lules cancer\u00edgenes <\/strong><\/p>\n

Aquest estudi se centra en la inhibici\u00f3 de la prote\u00efna fosfatasa 2A (PP2A), un enzim que desenvolupa un paper crucial en la regulaci\u00f3 de m\u00faltiples vies de senyalitzaci\u00f3 cel\u00b7lular relacionades amb el c\u00e0ncer. L\u2019equip d\u2019investigaci\u00f3 ha descobert que la inhibici\u00f3 de PP2A no nom\u00e9s activa m\u00faltiples vies oncog\u00e8niques (hiperactivaci\u00f3), sin\u00f3 que tamb\u00e9 desencadena respostes d\u2019\u201destr\u00e8s\u201d a les c\u00e8l\u00b7lules cancer\u00edgenes.<\/p>\n

Mitjan\u00e7ant screenings gen\u00e8tics i de llibreries de compostos van identificar que la inhibici\u00f3 combinada de PP2A i WEE1, una quinasa implicada en la regulaci\u00f3 del cicle cel\u00b7lular,\u00a0 tenia un efecte antitumoral sin\u00e8rgic en m\u00faltiples models de c\u00e0ncer,\u00a0 aconseguint suprimir el creixement tumoral a nivell precl\u00ednic, tant en cultius cel\u00b7lulars com en models animals. A m\u00e9s, s\u2019ha observat que les c\u00e8l\u00b7lules cancer\u00edgenes tractades amb aquesta combinaci\u00f3 terap\u00e8utica no van desenvolupar resist\u00e8ncia, perdent capacitat per a formar tumors.<\/p>\n

Per l\u2019investigador del Programa ProCURE de l\u2019ICO i l\u2019IDIBELL, Alberto Villanueva<\/a>, \u201caquest descobriment, una vegada validat en assaigs cl\u00ednics amb pacients, pot representar un pas important cap el desenvolupament d\u2019un tractament efectiu per a pacients amb c\u00e0ncer de c\u00f2lon\u201d, i afegeix: \u201caquesta nova estrat\u00e8gia farmacol\u00f2gica, basada en la hiperactivaci\u00f3 de les vies de senyalitzaci\u00f3 oncog\u00e8niques, que provoca un gran estr\u00e8s cel\u00b7lular, combinada amb la disrupci\u00f3 de les vies de resposta cel\u00b7lular a l\u2019estr\u00e8s, tamb\u00e9 pot ser efectiva per al tractament d’altres tipus de tumors com es demostra en el treball.\u201d<\/p>\n

La inhibici\u00f3 combinada de les prote\u00efnes PP2A i WEE1 produeix un col\u00b7lapse en la replicaci\u00f3 de l\u2019ADN desencadenant l\u2019aparici\u00f3 de mitosi prematura seguida de \u201cmort cel\u00b7lular\u201d. Com a conseq\u00fc\u00e8ncia d’aquest treball en uns mesos comen\u00e7ar\u00e0 un assaig cl\u00ednic fase I a Holanda (NCT06109883) on s’avaluar\u00e0 la seguretat, la tolerabilitat, la farmacocin\u00e8tica i l\u2019efic\u00e0cia preliminar de la combinaci\u00f3 en pacients amb c\u00e0ncer colorectal metast\u00e0tic.<\/p>\n","protected":false},"excerpt":{"rendered":"
