Fins ara, la majoria d\u2019estudis gen\u00e8tics en recerca oncol\u00f2gica havien concentrat esfor\u00e7os en investigar les alteracions que tenen un efecte directe en els gens que codifiquen per prote\u00efnes. Tot i aix\u00ed, es creu que al voltant del 90% de les variants gen\u00e8tiques relacionades amb c\u00e0ncer es troben en les regions no codificants, les quals poden regular i afectar indirectament l\u2019activitat dels gens. Aquestes no s\u2019han estudiat tant perqu\u00e8, com que no codifiquen per prote\u00efnes, \u00e9s molt dif\u00edcil determinar com influencien i afecten el progr\u00e9s de la malaltia. En aquest sentit, es mantenen com un aut\u00e8ntic misteri per a la recerca actual.<\/p>\n

Per\u00f2 ara, un equip investigadors del Shenzhen Bay Laboratory (SZBL) liderat pel Dr. Lei Li, en col\u00b7laboraci\u00f3 amb el grup de Regulaci\u00f3 G\u00e8nica de la Identitat Cel\u00b7lular<\/a> de l\u2019IDIBELL dirigit per la Dra. Mireya Plass, ha fet un pas m\u00e9s per tractar de comprendre l\u2019impacte d\u2019aquestes regions en el risc i desenvolupament de c\u00e0ncer. Aquesta col\u00b7laboraci\u00f3 ha resultat en una publicaci\u00f3 a la revista Nature Communications<\/a>, on la primera autora \u00e9s Hui Chen, estudiant de doctorat fruit del programa conjunt entre ambdues institucions.<\/p>\n

A l\u2019estudi, han analitzat el rol crucial de variants trobades en regions no codificants, fixant-se especialment en les que participen en un proc\u00e9s bioqu\u00edmic anomenat APA (poliadenilaci\u00f3 alternativa, per les sigles en angl\u00e8s). En l\u2019estudi, han identificat centenars d\u2019aquestes variants involucrades en processos de creixement tumoral i mort cel\u00b7lular, fet que implicaria un rol determinant en el desenvolupament de c\u00e0ncer. La majoria d’elles s\u2019han associat amb c\u00e0ncer de mama i pr\u00f2stata, i el 62% no s\u2019havien trobat en estudis anteriors. Que un percentatge tan elevat fossin troballes completament noves demostra el potencial que t\u00e9 l\u2019an\u00e0lisi d\u2019aquestes variants per descobrir nous factors de risc per c\u00e0ncer, i revela nous mecanismes pels quals la gen\u00e8tica pot contribuir al debut i el desenvolupament cancer\u00f3s.<\/p>\n

Per focalitzar l\u2019estudi i validar-ne la utilitat amb un cas espec\u00edfic, l\u2019equip d\u2019investigadors s\u2019ha centrat en l\u2019an\u00e0lisi de les variants que afecten el gen CRLS1<\/em><\/span>, particularment relacionat amb c\u00e0ncer de mama. Com esperaven, han corroborat que les variants que alteren el proc\u00e9s d\u2019APA impacten en l\u2019abund\u00e0ncia de prote\u00efna que deriva de CRLS1<\/em><\/span>, augmentant-ne significativament la quantitat, fet que s\u2019ha vinculat amb un increment del risc de c\u00e0ncer de mama.<\/p>\n

En definitiva, es pret\u00e9n continuar amb l\u2019estudi de les noves variants identificades per entendre millor la seva implicaci\u00f3 en c\u00e0ncer i els mecanismes pels quals actuen. Indagar en aquests factors permetria, en un futur, poder plantejar estrat\u00e8gies terap\u00e8utiques en qu\u00e8 la diana fossin els processos APA.<\/p>\n
