La leuc\u00e8mia limf\u00e0tica cr\u00f2nica es caracteritza per la proliferaci\u00f3 de c\u00e8l\u00b7lules B del sistema immunitari. Tot i que sol ser un tipus de leuc\u00e8mia indolent, entre el 5% i el 10% dels pacients acaben desenvolupant un limfoma m\u00e9s agressiu i amb mal pron\u00f2stic. Aquest proc\u00e9s s’anomena la transformaci\u00f3 de Richter<\/strong>.<\/p>\n

Ara, un equip del Cl\u00ednic-IDIBAPS amb la participaci\u00f3 de l\u2019IDIBELL ha generat un model de ratol\u00ed que recrea aquesta transformaci\u00f3 i que permetr\u00e0 buscar nous tractaments per aquests pacients, que en aquests moments disposen de poques opcions terap\u00e8utiques. El grup investigador ha presentat tots els detalls d’aquest nou model \u00fanic a la revista cient\u00edfica Leukemia<\/a>.<\/p>\n

Per generar el model, han injectat c\u00e8l\u00b7lules d’un pacient amb leuc\u00e8mia limf\u00e0tica cr\u00f2nica a un ratol\u00ed. Aquest tipus de models s’anomenen xenoinjerts derivats de pacients<\/strong>, i \u00e9s el primer descrit en el que a partir de c\u00e8l\u00b7lules d\u2019un pacient amb leuc\u00e8mia limf\u00e0tica cr\u00f2nica es genera un model de transformaci\u00f3 de Richter, recapitulant el que passa en el pacient mesos despr\u00e9s. Segonse els investigadors, aquest descobriment \u00e9s una eina primordial per entendre aquesta transformaci\u00f3 i buscar noves ter\u00e0pies m\u00e9s efectives<\/strong>.<\/p>\n

Un tractament prometedor<\/strong><\/p>\n

Les an\u00e0lisis moleculars que s’han pogut realitzar gr\u00e0cies a aquest nou model han desvelat que, un cop desenvolupada la transformaci\u00f3 de Richter, les c\u00e8l\u00b7lules del limfoma mostren una alta fosforilaci\u00f3 oxidativa i una baixa senyalitzaci\u00f3 del receptor de les c\u00e8l\u00b7lules B, el que explicaria la resist\u00e8ncia a certs tractaments que desenvolupen molts pacients.<\/p>\n

Arran d’aquests resultats, els investigadors van testar si la inhibici\u00f3 de la fosforilaci\u00f3 oxidativa podia aturar o alentir l’evoluci\u00f3 del limfoma. Els resultats obtinguts, tan en c\u00e8l\u00b7lules en cultiu com en un model de ratol\u00ed, s\u00f3n prometedors, ja que es mostra una reducci\u00f3 significativa de la proliferaci\u00f3 de les c\u00e8l\u00b7lules tumorals<\/strong>.<\/p>\n

Aquest projecte ha estat finan\u00e7at pel Ministeri de Ci\u00e8ncia i Innovaci\u00f3, pels Fons de Desenvolupament Regional Europeus, pel programa CERCA, el CIBERONC, el Ministeri d’Universitats, i les ajudes de l’American Association for Cancer Research, la European Hematology Association, i la Lady Tata Memorial Trust.<\/p>\n
