El proc\u00e9s pel qual es formen els \u00f2rgans, passant d’un estat immadur a un completament madur i funcional, \u00e9s extremadament complex i desconegut, encara hi ha molts mecanismes cel\u00b7lulars i moleculars que controlen el proc\u00e9s que s\u00f3n desconeguts. En el cas de l’intest\u00ed, aix\u00f2 implica adquirir la capacitat de digerir i absorbir els nutrients, proc\u00e9s controlat molecularment per factors de transcripci\u00f3 espec\u00edfics. Aquests factors de transcripci\u00f3 s\u00f3n els responsables d’activar i desactivar els gens que codifiquen prote\u00efnes que permeten a les c\u00e8l\u00b7lules transitar entre c\u00e8l\u00b7lules mare fetals o adultes i c\u00e8l\u00b7lules diferenciades.<\/p>\n

Desentrellar el funcionament de determinats factors de transcripci\u00f3<\/strong> en la formaci\u00f3 de l’intest\u00ed ha estat l’objectiu de l’equip del Professor Kim Bak Jensen<\/strong>, de la Novo Nordisk Foundation of Stem Cell Medicine (reNEW), en el que tamb\u00e9 hi ha participat el Dr. Jordi Guiu<\/strong>, cap del grup de recerca en Plasticitat i Regeneraci\u00f3 Cel\u00b7lular<\/a> de l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) i del Programa de Translaci\u00f3 Cl\u00ednica de Medicina Regenerativa de Catalunya (P-CMR[C]). Ara els seus resultats es presenten en dos articles publicats a la revista cient\u00edfica Science Advances<\/em><\/a>.<\/p>\n

En els dos estudis publicats, l’equip cient\u00edfic utilitza com a model l\u2019organoid d’intest\u00ed<\/strong>, una estructura tridimensional similars a miniintestins creada al laboratori a partir de c\u00e8l\u00b7lules mare. Gr\u00e0cies a la utilitzaci\u00f3 de diferents aproximacions moleculars demostren la utilitat dels models d’organoids per identificar els factors que regulen el dest\u00ed cel\u00b7lular, i revela que els factors de transcripci\u00f3 SMARCA4 i SMARCC1 eviten la diferenciaci\u00f3 preco\u00e7 de les c\u00e8l\u00b7lules mare durant el desenvolupament intestinal. A m\u00e9s, identifiquen l’activitat transcripcional de la prote\u00efna YAP com a principal reguladora de l’estat fetal immadur.<\/p>\n

El Dr. Jordi Guiu<\/strong> explica: “Entendre la transici\u00f3 d’un \u00f2rgan immadur a un \u00f2rgan madur ens podria ajudar a con\u00e8ixer com podem entrenar c\u00e8l\u00b7lules cultivades al laboratori per transitar en diferents estadis de maduresa. Aquest coneixement en \u00faltima inst\u00e0ncia ajudaria a generar ter\u00e0pies cel\u00b7lulars, per exemple, mitjan\u00e7ant la producci\u00f3 de c\u00e8l\u00b7lules epitelials al laboratori que es podrien trasplantar a pacients amb ulceres intestinals com poden ser el pacients amb la malaltia de Chron, o pacients tractats amb radioter\u00e0pia a la cavitat abdominal, entre d\u2019altres defectes epitelials<\/em>“.<\/p>\n

L\u2019Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) \u00e9s un centre de recerca en biomedicina creat l\u2019any 2004. L’any 2009 es va convertir en un dels cinc primers centres d\u2019investigaci\u00f3 espanyols acreditats com a institut d\u2019investigaci\u00f3 sanit\u00e0ria per l\u2019Instituto de Salud Carlos III.
