Mentre que la malaltia de Parkinson afecta el 3% de la poblaci\u00f3 major de 65 anys, i l’edat mitjana d’aparici\u00f3 s\u00f3n 60 anys. El Parkinson juvenil, que representa el 10% de tots els casos de Parkinson, s’inicia abans dels 40 anys. Ara, un grup d’investigadors de l’Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL), la Universitat de Barcelona (UB) i l’Institut de Neuroci\u00e8ncies (UBNeuro) ha desxifrat per primera vegada el mecanisme molecular pel qual aquesta mutaci\u00f3 s\u2019associa al Parkinson juvenil.<\/p>\n

L’equip, liderat pel Dr. Francisco Ciruela<\/strong>, s’ha centrat a estudiar l’impacte mecan\u00edstic que t\u00e9 la mutaci\u00f3 del receptor cerebral pr\u00e8viament definida com a possible causa de la malaltia primerenca. Els resultats, presentats a la revista Biomedicine and Pharmacotherapy<\/em><\/a>, demostren que la mutaci\u00f3 redueix la capacitat d\u2019aquest receptor per interaccionar amb d\u2019altres receptors d\u2019adenosina<\/strong>, concretament amb el receptor tipus 2, el que provocaria un increment en l\u2019excitabilitat dels circuits neuronals a la regi\u00f3 del cervell anomenada cos estriat.<\/p>\n

Nosaltres postulem que la incapacitat d\u2019interaccionar d\u2019ambd\u00f3s receptors d\u2019adenosina generaria hiperexcitabilitat glutamat\u00e8rgica als circuits neuronals de l\u2019estriat, mecanisme clau en la patog\u00e8nia del Parkinson juvenil<\/em>“, afirma el Dr. Ciruela<\/strong>, cap del grup de recerca en Neurofarmacologia i Dolor de l’IDIBELL, i catedr\u00e0tic de la Facultat de la Medicina i Ci\u00e8ncies de la Salut de la Universitat de Barcelona.<\/p>\n

Desequilibri en l’excitaci\u00f3 del circuit neuronal<\/h3>\n

La mutaci\u00f3 estudiada afecta al receptor d’adenosina tipus 1. Aquest receptor exerceix un efecte inhibidor sobre el seu hom\u00f2leg, el receptor d’adenosina tipus 2, que facilita l\u2019alliberaci\u00f3 de glutamat, i l\u2019excitabilitat del circuit. Per tant, segons els resultats publicats, la mutaci\u00f3 impediria la interacci\u00f3 molecular i funcional d\u2019ambd\u00f3s receptors d\u2019adenosina, i en conseq\u00fc\u00e8ncia, facilitaria l\u2019alliberaci\u00f3 de glutamat, que provocaria una hiperexcitabilitat als circuits neuronals de l\u2019estriat.<\/p>\n

Els receptors d’adenosina, s\u00f3n receptors cerebrals acoblats a prote\u00efnes G i involucrats en funcions motores. Pr\u00e8viament, ja s’havia apuntat la seva implicaci\u00f3 en malalties neurodegeneratives com el Parkinson.<\/p>\n

En aquest estudi tamb\u00e9 hi han participat investigadors i investigadores de la Universitat Aut\u00f2noma de Barcelona, del Luxembourg Institute of Health (LIH), i del National Institutes of Health de Baltimore.<\/p>\n
