ha aconseguit identificar els mecanismes moleculars regulats per aquesta prote\u00efna i ha desxifrat el paper protector contra el c\u00e0ncer<\/strong>.<\/p>\n“Si b\u00e9 les c\u00e8l\u00b7lules canceroses del fetge tenen especial avidesa per la glucosa i els l\u00edpids, cosa que els permet multiplicar-se r\u00e0pidament, NOX4 frena aquests processos<\/em>“, explica Fabregat<\/strong>, que tamb\u00e9 \u00e9s professora de la Universitat de Barcelona i cap de grup del CIBER a l’\u00c0rea de Malalties Hep\u00e0tiques i Digestives (CIBEREHD). Afegeix: “Quan l’activitat de NOX4 \u00e9s baixa, es reprograma el metabolisme cel\u00b7lular i es provoca un estr\u00e8s oxidatiu, cosa que afavoreix que el tumor proliferi m\u00e9s i augmenti la seva capacitat d’envair altres teixits. Per tant, els nivells de NOX4 poden condicionar l’evoluci\u00f3 del c\u00e0ncer de fetge<\/em>“.<\/p>\n <\/p>\n
Difer\u00e8ncies entre prote\u00efnes NOX<\/h3>\n
Durant l’aparici\u00f3 del c\u00e0ncer de fetge es crea un microambient oxidatiu que provoca estr\u00e8s cel\u00b7lular, cosa que activa les prote\u00efnes NOX. No obstant aix\u00f2, no totes les prote\u00efnes de la fam\u00edlia tenen les mateixes funcions. Mentre que NOX4 t\u00e9 un efecte protector, tal com ha revelat aquest estudi, NOX1 contribueix a la progressi\u00f3 c\u00e0ncer. “Els resultats apunten que NOX1 afavoreix que s’incrementi encara m\u00e9s l’estr\u00e8s oxidatiu, proc\u00e9s que NOX4 aconsegueix reduir<\/em>“, explica Irene Pe\u00f1uelas<\/strong>, investigadora postdoctoral a l’IDIBELL, aix\u00ed com al CIBEREHD i primera signant de l’estudi.<\/p>\nLes prote\u00efnes NOX estarien assolint aquests canvis mitjan\u00e7ant la regulaci\u00f3 dels factors de transcripci\u00f3 MYC i NRF2, responsables de l’expressi\u00f3 de molts gens implicats en el c\u00e0ncer. “Hem vist que en abs\u00e8ncia de NOX4 s’augmenta l’expressi\u00f3 de NOX1 o NOX2, cosa que provoca un augment en l’activitat de NRF2 i MYC, que s\u00f3n responsables de canvis als mitocondris i una reprogramaci\u00f3 del metabolisme oxidatiu, augmentant la capacitat energ\u00e8tica de la c\u00e8l\u00b7lula<\/em>“, indica Pe\u00f1uelas<\/strong>.<\/p>\nDe fet, estudis previs del mateix grup havien assenyalat que l’eliminaci\u00f3 de NOX4 estimula la proliferaci\u00f3 de les c\u00e8l\u00b7lules del fetge. “Les aplicacions terap\u00e8utiques de NOX4 s\u00f3n molt \u00e0mplies. Ens pot interessar inhibir-la per promoure la regeneraci\u00f3 del fetge en situacions de malalties cr\u00f2niques que produeixen fibrosi hep\u00e0tica, o justament el contrari, compensar la seva manca d’activitat en casos de c\u00e0ncer per frenar la progressi\u00f3<\/em>“, conclou Fabregat<\/strong>.<\/p>\nEl treball s’ha realitzat en el context del consorci CIBER pertanyent a l’Institut de Salut Carlos III i ha estat liderat des de l’IDIBELL amb la participaci\u00f3 de professionals de l’Institut Catal\u00e0 d’Oncologia, l’Hospital Universitari de Bellvitge i la Universitat de Barcelona en col\u00b7laboraci\u00f3 amb grups d’investigaci\u00f3 a escala nacional del CIC bioGUNE, el Centre de Biologia Molecular Severo Ochoa, la Universitat Aut\u00f2noma de Madrid, l’IRB Barcelona, i a escala internacional amb cient\u00edfics de el University College Dublin.<\/p>\n
