Aquesta nova eina demostra ser efica\u00e7 i especifica en el model de cuc Caernorhabditis elegans i al peix zebra (zebrafish).<\/p>\nEl Dr. Juli\u00e1n Cer\u00f3n<\/strong>, cap grup a l’IDIBELL i col\u00edder del projecte juntament amb Miguel \u00c1ngel Moreno-Mateos<\/strong> del CABD, declara: “Aquest nou sistema expandeix les possibilitats de l’edici\u00f3 gen\u00e8tica, podent arribar a moltes m\u00e9s zones del genoma fins ara molt dif\u00edcils de modificar<\/em>“.<\/p>\nPer la seva banda, Jeremy Vicencio<\/strong>, investigador de l’IDIBELL i primer autor del treball juntament amb Carlos S\u00e1nchez-Bola\u00f1os<\/strong> del CABD, afirma que “ha estat molt emocionant trobar les condicions en paral\u00b7lel en cucs i peixos on aquest nou CRISPR-Cas9 funcion\u00e9s tan b\u00e9<\/em>“.<\/p>\n <\/p>\n
Condicions per al seu funcionament en animals<\/h3>\n
La modificaci\u00f3 del domini de reconeixement de l’ADN d’aquest nou sistema millorat de CRISPR-Cas9 va ser ideat i executat per experts en estructura de prote\u00efnes del laboratori de Ben Kleinstiver a Boston, que tamb\u00e9 han participat en l’estudi. Fins ara, aquesta nova eina havia mostrat efic\u00e0cia en l\u00ednies cel\u00b7lulars i plantes, per\u00f2 no s’havia aconseguit aplicar en l’edici\u00f3 del genoma d’animals.<\/p>\n
En aquest treball, els laboratoris del Dr. Cer\u00f3n a IDIBELL i del Dr. Moreno-Mateo a CABD han unit forces per fer funcionar aquesta t\u00e8cnica en animals. Tot i que en un inici el nou CRISPR-Cas9 semblava no ser \u00fatil per al genoma animal, els investigadors van aconseguir fer-lo funcionar incrementant la seva concentraci\u00f3 fins a sis vegades<\/strong>.<\/p>\n“Encara que no ho sabem amb certesa, per\u00f2 \u00e9s possible que la necessitat d’incrementar la concentraci\u00f3 sigui deguda al fet que aquest nou sistema t\u00e9 m\u00e9s potencials dianes d’acci\u00f3 i, per tant, necessiti m\u00e9s prote\u00efnes Cas9 que busquin en lloc adequat<\/em> -comenta el Dr. Cer\u00f3n<\/strong>-.
