La quimioter\u00e0pia amb cisplat\u00ed \u00e9s la m\u00e9s comuna per abordar els tumors s\u00f2lids. No obstant aix\u00f2, malgrat la seva efic\u00e0cia, els efectes adversos associats, especialment el potencial neurot\u00f2xic, i les resist\u00e8ncies al tractament que poden desenvolupar les c\u00e8l\u00b7lules tumorals fan augmentar el frac\u00e0s de la ter\u00e0pia i, en conseq\u00fc\u00e8ncia, la mortalitat.<\/p>\n

Ara un grup d’investigadors de l’IDIBELL i de l’ICO han fet servir un model de cuc per estudiar els mecanismes d’adquisici\u00f3 de resist\u00e8ncia al tractament i cercar noves vies per evitar-ho<\/strong>.<\/p>\n

El treball, publicat a la revista Disease Models & Mechanisms<\/em><\/a>, ha estat liderat pel Dr. Juli\u00e1n Cer\u00f3n<\/strong>, de l’IDIBELL, i el Dr. Ernest Nadal<\/strong>, de l’IDIBELL i de l’ICO, amb la col\u00b7laboraci\u00f3 del Dr. Alberto Villanueva<\/strong>, de la mateixa instituci\u00f3. En ell es descriu que una dieta rica en glucosa fa els cucs m\u00e9s sensibles al cisplat\u00ed. A m\u00e9s, mostren que el tractament amb dopamina protegeix de l’efecte neurot\u00f2xic de la quimioter\u00e0pia al model animal.<\/p>\n

Moltes vegades els onc\u00f2legs es veuen obligats a reduir la dosi o fins i tot aturar el tractament amb cisplat\u00ed a causa de la seva toxicitat. “Els nostres resultats suggereixen explorar l’\u00fas de dopamina o derivats per mitigar els efectes neurot\u00f2xics d’aquesta quimioter\u00e0pia. Aix\u00f2 permetria mantenir les dosis desitjades de cisplat\u00ed i augmentar l’efic\u00e0cia del tractament contra els tumors s\u00f2lids<\/em>“, afirma la Dra. Carmen Mart\u00ednez-Fern\u00e1ndez<\/strong>, primera autora del treball.<\/p>\n

El cuc per estudiar una malaltia humana<\/h3>\n

El cuc de terra C. elegans<\/em> \u00e9s \u00e0mpliament utilitzat en la investigaci\u00f3 biom\u00e8dica perqu\u00e8, tot i ser molt simple i petit, t\u00e9 la majoria de les estructures dels animals m\u00e9s complexos i comparteix moltes de les vies metab\u00f2liques. A m\u00e9s, un 40% del seu genoma compr\u00e8n gens hom\u00f2legs als humans.<\/p>\n

La manipulaci\u00f3 del genoma per la tecnologia CRISPR en aquest organisme permet crear models per estudiar malalties m\u00e9s r\u00e0pidament que altres models animals. Tot plegat, unit a altres trets com la transpar\u00e8ncia del seu cos, un curt cicle de vida, i un baix cost de manteniment, el converteix en una eina ideal per a la investigaci\u00f3 biom\u00e8dica.<\/p>\n

Les caracter\u00edstiques del model C. elegans ens han perm\u00e8s investigar en un organisme pluricel\u00b7lular la resposta al tractament amb cisplat\u00ed, cosa que ens aporta una visi\u00f3 m\u00e9s global i sist\u00e8mica dels seus efectes<\/em>“, afirma el Dr. Cer\u00f3n<\/strong>.<\/p>\n

L\u2019Institut d’Investigaci\u00f3 Biom\u00e8dica de Bellvitge (IDIBELL) \u00e9s un centre de recerca en biomedicina creat l\u2019any 2004. A m\u00e9s, forma part del programa \u201cHR Excellence in Research\u201d de la Uni\u00f3 Europea i \u00e9s membre de EATRIS i REGIC.
