Davant d’una situaci\u00f3 de manca de nutrients, oxigen i energia, les c\u00e8l\u00b7lules tumorals paren de cr\u00e9ixer i creen un petit reservori amb tot el material necessari per reiniciar r\u00e0pidament el creixement i la divisi\u00f3 cel\u00b7lular tan bon punt la situaci\u00f3 torni a ser favorable. Aquesta \u00e9s la principal conclusi\u00f3 d’un treball publicat a la revista Science Advances<\/em><\/a> del grup de recerca en Metabolisme i C\u00e0ncer<\/a> de l’Institut de Recerca Biom\u00e8dica de Bellvitge (IDIBELL) i la Universitat de Barcelona (UB), i finan\u00e7ada per la Associaci\u00f3 Espanyola Contra el C\u00e0ncer.<\/p>\n

El c\u00e0ncer es produeix per la proliferaci\u00f3 descontrolada de c\u00e8l\u00b7lules, que acaben envaint i destruint teixits i \u00f2rgans. Per aquesta ra\u00f3, perqu\u00e8 un tumor augmenti de mida \u00e9s imprescindible que les c\u00e8l\u00b7lules tumorals produeixin totes les prote\u00efnes necess\u00e0ries per crear la biomassa requerida per cr\u00e9ixer i dividir-se. Les f\u00e0briques que produeixen les prote\u00efnes s\u00f3n els ribosomes, per aix\u00f2 la capacitat de generar nous ribosomes \u00e9s clau en els tumors m\u00e9s agressius.<\/p>\n

L’estudi liderat pel Dr. Antonio Gentilella, investigador principal de l’IDIBELL i professor de la UB, descriu que, quan les c\u00e8l\u00b7lules tumorals es troben en un ambient baix en nutrients, s\u00f3n capaces de detectar-ho i enviar un senyal intern perqu\u00e8 la prote\u00efna LARP1 i la subunitat ribosomal 40S segrestin tots els ARNm necessaris per formar ribosomes.<\/p>\n

\u201cEn una situaci\u00f3 desfavorable la c\u00e8l\u00b7lula degrada tot el que pot per aconseguir l’energia i els recursos que li permetin sobreviure en mode de baix consum i d\u2019estalvi d’energia. Aquesta caixa forta d’informaci\u00f3 per produir ribosomes permet guardar-los i protegir-los de la degradaci\u00f3 per despr\u00e9s poder-los generar r\u00e0pidament quan la situaci\u00f3 torni a la normalitat i la c\u00e8l\u00b7lula pugui tornar a cr\u00e9ixer. Pr\u00e0cticament Ribosomes estil IKEA<\/em>\u201d, comenta el Dr. Antonio Gentilella<\/strong>, l\u00edder del projecte<\/p>\n

Pedro Fuentes<\/strong> i Joffrey Pelletier<\/strong>, tots dos investigadors de l’IDIBELL i primers autors de l’article, cometin que es tracta d’un mecanisme de resist\u00e8ncia metab\u00f2lica observat en molts tipus tumorals, des de c\u00e8l\u00b7lules de c\u00e0ncer de c\u00f2lon i recte, de pulm\u00f3 o de bufeta, i potencialment molts altres tumors.<\/p>\n

Podria el reservori de ribosomes influir en la reaparici\u00f3 de tumors?<\/h4>\n

L’ambient de creixement dels tumors, aix\u00ed com la quimioter\u00e0pia, creen les condicions desfavorables de manca de nutrients i oxigen que podrien promoure la creaci\u00f3 de reservoris de ribosomes en les c\u00e8l\u00b7lules que sobreviuen a aquestes condicions. Per tant, \u00e9s f\u00e0cil pensar que aquest pugui constituir un mecanisme important per a la reaparici\u00f3 dels tumors despr\u00e9s de la ter\u00e0pia.<\/p>\n

\u201cLa quimioter\u00e0pia pot reduir molt la mida dels tumors, per\u00f2 qu\u00e8 queda al final? \u00c9s molt probable que el mateix tractament hagi seleccionat les c\u00e8l\u00b7lules que per una banda hagin baixat el consum energ\u00e8tic i alhora hagin generat un reservori de ribosomes i, per tant, els \u00e9s m\u00e9s f\u00e0cil tant sobreviure com tornar a cr\u00e9ixer despr\u00e9s del tractament\u201d,<\/em> comenta el Dr. Gentilella. \u201cEn aquests moments estem estudiant a nivell cl\u00ednic en quin grau aix\u00f2 est\u00e0 passant en diferents tipus de tumors i si som capa\u00e7os de parar-ho per evitar-ne la reaparici\u00f3\u201d.<\/em><\/p>\n

Aquest treball \u00e9s la continuaci\u00f3 d\u2019uns quants anys de recerca del grup del Dr. Gentilella i deu molt a la contribuci\u00f3 intel\u00b7lectual del Dr. George Thomas, reconegut expert mundial en ribosomes i investigador em\u00e8rit de l’IDIBELL.<\/p>\n

L’any 2009 es va convertir en un dels cinc primers centres d\u2019investigaci\u00f3 espanyols acreditats com a institut d\u2019investigaci\u00f3 sanit\u00e0ria per l\u2019Instituto de Salud Carlos III.
