En les c\u00e8l\u00b7lules femenines, l’expressi\u00f3 del cromosoma X se silencia per equilibrar l’expressi\u00f3 g\u00e8nica entre les dones XX i homes XY. Un dels mecanismes m\u00e9s generals per a la regulaci\u00f3 de l’expressi\u00f3 g\u00e8nica \u00e9s la distribuci\u00f3 de l’ADN dins el nucli. En les c\u00e8l\u00b7lules de mam\u00edfers, distingim dos compartiments: eucromatina, situada al centre del nucli, on l’ADN est\u00e0 m\u00e9s relaxat i els gens presenten una alta taxa d’expressi\u00f3; i heterocromatina, a la perif\u00e8ria del nucli amb ADN compactat i l’expressi\u00f3 g\u00e8nica silenciada.<\/p>\n

Un estudi dirigit per la Dra. Maria Carolina Florian<\/strong>, del Programa de Medicina Regenerativa de l’Institut de Recerca Biom\u00e8dica de Bellvitge (IDIBELL<\/a>), i el Dr. Medhanie A. Mullaw<\/strong>, de l’Hospital Universitari d’Ulm, ha demostrat que la prote\u00efna nuclear Lamin A\/C crea un compartiment en el nucli de les c\u00e8l\u00b7lules mare joves de la medul\u00b7la \u00f2ssia per inactivar el cromosoma X en les c\u00e8l\u00b7lules femenines.<\/p>\n

L’estudi, publicat a Stem Cell Report<\/em><\/a>, indica que en les c\u00e8l\u00b7lules mare de la medul\u00b7la \u00f2ssia envellides, el compartiment d’inactivaci\u00f3 de Lamin A\/C<\/strong> es redueix en gran manera, el que altera la localitzaci\u00f3 i inactivaci\u00f3 de la perif\u00e8ria del cromosoma X. Les observacions indiquen una relocalitzaci\u00f3 general del cromosoma X “inactivat” a regions m\u00e9s actives del nucli, el que es correlaciona amb la major expressi\u00f3 g\u00e8nica d’aquests cromosomes en c\u00e8l\u00b7lules envellides.<\/p>\n

La desregulaci\u00f3 de la inactivaci\u00f3 del cromosoma X est\u00e0 fortament associada a la s\u00edndrome mielodispl\u00e0stica<\/strong>, un grup de c\u00e0ncers que afecta amb m\u00e9s freq\u00fc\u00e8ncia a la gent gran, en el qual les c\u00e8l\u00b7lules sangu\u00ednies no maduren. Per tant, l’exist\u00e8ncia d’una alteraci\u00f3 dependent de l’edat de la inactivaci\u00f3 del cromosoma X pot tenir implicacions importants per a comprendre la fisiologia de l’hematopoesi durant l’envelliment i la patog\u00e8nesi de les neopl\u00e0sies hematopo\u00e8tiques relacionades amb l’edat.<\/p>\n
