Els nostres interessos de recerca es troben principalment a l’\u00e0rea de les c\u00e8l\u00b7lules mare i la medicina regenerativa. En general, el nostre objectiu d’investigaci\u00f3 \u00e9s aprofitar les tecnologies emergents de c\u00e8l\u00b7lules mare per comprendre millor les malalties neurodegeneratives, com ara la MP i les malalties del neurodesenvolupament, com la THD, i desenvolupar eines noves per al descobriment de mecanismes i tractaments de malalties. Durant els darrers 10 anys, en estreta col\u00b7laboraci\u00f3 amb el grup del Dr. Angel Raya i el Dr. Eduardo Tolosa, hem acumulat una gran col\u00b7lecci\u00f3 de l\u00ednies iPSC que representen pacients amb malaltia de Parkinson (MP), incloses formes idiop\u00e0tiques i gen\u00e8tiques associades a la mutaci\u00f3 LRRK2G2019S. , aix\u00ed com familiars portadors asimptom\u00e0tics de LRRK2G2019S i individus sans. Per a la majoria dels pacients i portadors de LRRK2G2019S, el seu laboratori tamb\u00e9 va generar controls iPSC editats amb gens isog\u00e8nics. Aquestes l\u00ednies d’iPSC s’han utilitzat per generar neurones dopamin\u00e8rgiques que mostren les caracter\u00edstiques fenot\u00edpiques de la MP i recapitulen les alteracions cel\u00b7lulars, moleculars i epigen\u00e8tiques que es troben a les neurones dels pacients amb MP. A m\u00e9s, aprofitant els avantatges de la tecnologia de modelatge de malalties basada en iPSC, recentment revelem la contribuci\u00f3 pat\u00f2gena de les c\u00e8l\u00b7lules no neuronals a la neurodegeneraci\u00f3 dopamin\u00e8rgica a l’EP, descobrim alteracions funcionals primerenques a les xarxes neuronals derivades d’iPSC de la MP i analitzem les mol\u00e8cules petites que prevenen neurodegeneraci\u00f3 a la MP. Tamb\u00e9 adoptem enfocaments d’edici\u00f3 de gens als nostres models iPSC per determinar com els objectius espec\u00edfics influeixen en l’agregaci\u00f3 de prote\u00efnes i la fisiopatologia neuronal i per revelar els mecanismes pels quals les noves variants gen\u00e8tiques protectores afecten l’inici i la progressi\u00f3 de la malaltia. Recentment, tamb\u00e9 estem desenvolupant models in vitro experimentals avan\u00e7ats, com a organoides similars a cervells, per recapitular la complexitat del cervell hum\u00e0.<\/p>\n
