Crecimiento celular<\/strong>
\nEl crecimiento celular est\u00e1 relacionado con la cantidad de prote\u00ednas que sintetizan los ribosomas de la c\u00e9lula, una maquinaria intracelular encargada de traducir las mol\u00e9culas de ARN mensajero, proveniente del ADN, en cadenas de amino\u00e1cidos, formando las prote\u00ednas). El mal funcionamiento en la formaci\u00f3n de los ribosomas est\u00e1 relacionado con des\u00f3rdenes asociados al crecimiento celular aberrante como la anemia y el c\u00e1ncer. La activaci\u00f3n de p53 induce la activaci\u00f3n de un programa de muerte celular que previene que las c\u00e9lulas que crecen de forma aberrante puedan desarrollar un tumor. En condiciones normales, se mantiene en niveles bajos para evitar da\u00f1ar las c\u00e9lulas sanas. La prote\u00edna principal en el mantenimiento de niveles bajos de p53 es Hdm2, que en condiciones de crecimiento celular normal, degrada p53. Los ribosomas est\u00e1n formados por dos subunidades denominadas 40S y 60S. En la formaci\u00f3n de la subunidad 60S participan diversas mol\u00e9culas, incluyendo L5, L11 y 5S rRNA que forman un complejo pre-ribosomal antes de ser incorporado a la subunidad 60S.<\/p>\nEl laboratorio de George Thomas ha demostrado que cuando existe un da\u00f1o en los ribosomas, o cuando la formaci\u00f3n de ribosomas est\u00e1 hiperactivada el complejo pre-ribosomal l5\/L11\/5S rRNA se desv\u00eda de su ruta para formar ribosomas y se une a Hdm2 bloqueando su actividad, permitiendo que aumenten los niveles de p53 y la inducci\u00f3n de la muerte celular. Anteriormente, el equipo de George Thomas ya hab\u00eda demostrado que L5 y L11 regula Hdm2. Ahora han descubierto la existencia del este complejo pre-ribosomal formado por L5, L11 y tambi\u00e9n 5S rRNA y su papel como supresor de tumores. Estos resultados apuntan a un antiguo v\u00ednculo evolutivo entre biog\u00e9nesis de los ribosomas y el c\u00e1ncer<\/p>\n
George Thomas ha explicado que entender c\u00f3mo funciona y c\u00f3mo se regula p53 es muy importante ya que “m\u00e1s del 50% de los tumores presentan mutaciones en p53 o sobreexpresan Hdm2 o Hdm4, que bloquean la actividad de p53”.
