Investigadores del Instituto de Investigaci\u00f3n Biom\u00e9dica de Bellvitge (IDIBELL) y la Universidad de Barcelona han descubierto la interacci\u00f3n entre la prote\u00edna HERC2 con otra prote\u00edna llamada p53 que se encuentra inactiva en m\u00e1s de la mitad de los tumores en humanos. Los resultados del estudio se han publicado en la revista Journal of Biological Chemistry.

El equipo de Jos\u00e9 Luis Rosa, del grupo de investigaci\u00f3n en factores de crecimiento y diferenciaci\u00f3n celular del IDIBELL estudia los mecanismos moleculares de la familia de prote\u00ednas HERC. Estas prote\u00ednas son ubiquitina ligasas que regulan la funci\u00f3n de otras prote\u00ednas. \u00abEn los \u00faltimos a\u00f1os\u00bb ha explicado Rosa \u00abse ha demostrado que alteraciones de la familia de prote\u00ednas HERC est\u00e1n asociadas con diversas patolog\u00edas incluyendo ataxia, retraso mental y c\u00e1ncer\u00bb.
\u00abEn condiciones fisiol\u00f3gicas, la actividad de la prote\u00edna p53 protege a la c\u00e9lula de un crecimiento descontrolado. As\u00ed, mutaciones u otros mecanismos que inhiben la funci\u00f3n de p53, se asocian con un aumento de la proliferaci\u00f3n celular \u00abseg\u00fan dice el investigador\u00bb como un tumor se forma por un crecimiento celular descontrolado, no es de extra\u00f1ar que una etapa importante en la progresi\u00f3n tumoral sea la inactivaci\u00f3n de p53″.
La prote\u00edna p53 regula la expresi\u00f3n de genes involucrados en importantes procesos celulares como la proliferaci\u00f3n celular. La actividad de p53 depende de adquirir una estructura de cuatro mol\u00e9culas de p53 asociadas denominada tetr\u00e1mero. Los investigadores han descubierto que HERC2 es necesaria para la formaci\u00f3n del tetr\u00e1mero de p53 (proceso denominado oligomerizaci\u00f3n) y de esta forma HERC2 regula la actividad de p53 en condiciones fisiol\u00f3gicas.

El estudio sugiere que mutaciones en HERC2 tambi\u00e9n podr\u00edan estar asociadas a c\u00e1ncer en humanos. \u00abEn el laboratorio hemos observado que si a las c\u00e9lulas les inactivamos HERC2 aumenta la proliferaci\u00f3n celular.
