La met\u00e1stasis es responsable del 90% de las muertes en pacientes con c\u00e1ncer. Por ello, entender los mecanismos responsables de este proceso es uno de los principales objetivos de la investigaci\u00f3n en c\u00e1ncer. La met\u00e1stasis consiste en una serie de pasos encadenados en los que el tumor primario invade los tejidos vecinos y termina diseminando por todo el organismo. Uno de los tumores con mayor capacidad de producir met\u00e1stasis es el melanoma, que ha incrementado su incidencia en las \u00faltimas d\u00e9cadas, como consecuencia de la mayor exposici\u00f3n solar.
Hoy, un art\u00edculo publicado en la revista Nature Medicine dirigido por Manel Esteller, director del Programa de Epigen\u00e9tica y Biolog\u00eda del C\u00e1ncer (PEBC) del Instituto de Investigaci\u00f3n Biom\u00e9dica de Bellvitge (IDIBELL), investigador ICREA y profesor de gen\u00e9tica de la Universidad de Barcelona, \u200b\u200bdescribe un mecanismo que explica c\u00f3mo las c\u00e9lulas tumorales escapan de su lugar de origen haciendo met\u00e1stasis y, adem\u00e1s, pueden ser susceptibles a responder a nuevos tratamientos moleculares.
\u00abHemos comparado las c\u00e9lulas de los tumores originales de pacientes con las c\u00e9lulas que han hecho met\u00e1stasis y hemos visto que estas \u00faltimas presentan una alteraci\u00f3n espec\u00edfica en su material gen\u00e9tico. Concretamente, activan un gen que est\u00e1 silenciado tanto en las c\u00e9lulas sanas como en las c\u00e9lulas de los tumores primarios \u00abha explicado Esteller.
\u00abEste gen\u00bb contin\u00faa el investigador \u00abse enciende como una bombilla para guiar las c\u00e9lulas tumorales que escapan del tumor primario\u00bb.
El gen, llamado TBC1D16 encarga de modular oncogenes muy potentes como BRAF y EGFR y lo que es \u00abmuy interesante\u00bb afirma Esteller \u00abes que las met\u00e1stasis de melanoma se vuelven adictos ‘a estos oncogenes de manera que los f\u00e1rmacos que los tienen como diana, llamados inhibidores de BRAF y MEK son m\u00e1s efectivos \u00ab. Seg\u00fan explica Esteller \u00abestos estudios confirman que los tumores humanos cambian en su historia natural adapt\u00e1ndose a trav\u00e9s de la regulaci\u00f3n de su genoma (por cambios epigen\u00e9ticos) para sobrevivir en los nuevos tejidos donde hacen met\u00e1stasis. Pero a\u00fan m\u00e1s relevante, sugieren que el uso de tratamientos combinados que no permiten a la c\u00e9lula tumoral adaptarse son los m\u00e1s indicados para tratar la enfermedad \u00bb
