Investigadores del grupo de v\u00edas de se\u00f1alizaci\u00f3n y angiog\u00e9nesis del Instituto de Investigaci\u00f3n Biom\u00e9dica de Bellvitge (IDIBELL) liderados por Mariona Graupera han demostrado que la prote\u00edna PTEN es clave para evitar la proliferaci\u00f3n descontrolada de las c\u00e9lulas que forman los vasos sangu\u00edneos en el proceso de angiog\u00e9nesis. Los resultados del estudio se publican hoy en la revista Nature Communications.
El crecimiento de vasos sangu\u00edneos por brotes (sprouting en ingl\u00e9s) es un mecanismo central de la formaci\u00f3n de la vasculatura y comienza por la especializaci\u00f3n de c\u00e9lulas endoteliales en varios tipos de c\u00e9lulas. Por un lado, las c\u00e9lulas tip (punta en castellano) que gu\u00edan el sentido del crecimiento y que casi no proliferan y por otro, las c\u00e9lulas Stalk (tallo en castellano) que crecen detr\u00e1s para ir formando el lumen de los nuevos vasos sangu\u00edneos.
Estudios anteriores hab\u00edan demostrado que la ruta de se\u00f1alizaci\u00f3n de Notch est\u00e1 implicada en el control de las c\u00e9lulas Stalk bloqueando su proliferaci\u00f3n. El trabajo del equipo de Mariona Graupera demuestra como Notch activa PTEN y \u00e9sta promueve la parada del ciclo celular y por lo tanto evita la proliferaci\u00f3n de las c\u00e9lulas. Los investigadores han demostrado tanto en cultivos celulares como en ratones que en ausencia de PTEN, aunque Notch est\u00e9 activado, no se detiene la proliferaci\u00f3n de las c\u00e9lulas Stalk derivando en la formaci\u00f3n de una vasculatura aberrante.

\u00abEstudiar la fisiolog\u00eda de la angiog\u00e9nesis nos permite entender mejor las enfermedades en las que hay un crecimiento aberrante de los vasos sangu\u00edneos, como es el caso del c\u00e1ncer, y encontrar mejores aproximaciones terap\u00e9uticas\u00bb explic\u00f3 Mariona Graupera.
En los \u00faltimos a\u00f1os se ha visto que las terapias antiangiog\u00e9nicas son de corto recorrido y que a largo plazo pueden ser perjudiciales. Las aproximaciones actuales se dirigen a normalizar la vasculatura de modo que aunque los nutrientes y el ox\u00edgeno lleguen a las c\u00e9lulas tumorales tambi\u00e9n lo hagan los agentes quimioterap\u00e9uticos.
\u00abEn este sentido, se puede utilizar PTEN para normalizar la vasculatura y mantener la quiescencia, el equilibrio en la formaci\u00f3n desde vasos sangu\u00edneos\u00bb concluye la investigadora.
