Investigadores del grupo de Transformaci\u00f3n y Met\u00e1stasis del Instituto de Investigaci\u00f3n Biom\u00e9dica de Bellvitge (IDIBELL) liderados por Eva Gonz\u00e1lez Su\u00e1rez han descubierto una doble funci\u00f3n de la prote\u00edna RANK en la diferenciaci\u00f3n de los alveolos mamarios y la lactog\u00e9nesis. Los resultados se han publicado en la revista Stem Cells.
Hasta ahora se conoc\u00eda el papel positivo de la v\u00eda de RANK en la alveog\u00e9nesis (la diferenciaci\u00f3n de los alveolos mamarios) durante el principio de la gestaci\u00f3n. \u201cLo que hemos observado\u201d explica la investigadora Eva Gonz\u00e1lez-Su\u00e1rez \u201ces que en etapas posteriores del embarazo la activaci\u00f3n de RANK juega un papel negativo: inhibe la acci\u00f3n de la prolactina, impidiendo la fosforilaci\u00f3n de Stat5 y la lactog\u00e9nesis, la secreci\u00f3n de leche\u201d.
A trav\u00e9s de an\u00e1lisis de expresi\u00f3n g\u00e9nica, los investigadores han demostrado que la activaci\u00f3n de la v\u00eda de RANK a media gestaci\u00f3n impide la diferenciaci\u00f3n de las c\u00e9lulas alveolares en c\u00e9lulas secretoras de leche tanto en modelos de rat\u00f3n modificados gen\u00e9ticamente (con altos niveles de expresi\u00f3n de RANK) como en modelos de rat\u00f3n wild-type (sin ninguna modificaci\u00f3n gen\u00e9tica). Por otra parte, si a media gestaci\u00f3n inhibimos farmacol\u00f3gicamente la prote\u00edna RANKL en ratones wild type se produce una lactog\u00e9nesis precoz y exacerbada.
\u201cEstos resultados explican, a nivel molecular, el hecho de que tanto la ausencia como la sobreactivaci\u00f3n de RANK interfieran con la lactancia\u201d, explica Gonz\u00e1lez-Su\u00e1rez. Y a\u00f1ade que \u201cest\u00e1 claro que es necesaria una estricta regulaci\u00f3n de la v\u00eda de se\u00f1alizaci\u00f3n de RANK para que esta diferenciaci\u00f3n de la gl\u00e1ndula mamaria se realice de forma correcta\u201d.
