Investigadores del Grupo de Diferenciaci\u00f3n Celular del Programa de Epigen\u00e9tica y Biolog\u00eda del c\u00e1ncer (IDIBELL), liderados por la investigadora Maribel Parra, han identificado un represor transcripcional, la histona desacetilasa HDAC7, implicado en la generaci\u00f3n y la identidad de los linfocitos B, las c\u00e9lulas encargadas de crear anticuerpos en nuestro sistema inmunitario. Los resultados del estudio, publicado en The Journal of Experimental Medicine, demuestran que HDAC7 es necesaria para la formaci\u00f3n de linfocitos B.

El sistema hematopoy\u00e9tico comprende unos procesos complejos de diferenciaci\u00f3n celular para formar las diferentes c\u00e9lulas sangu\u00edneas. Dentro del sistema hematopoy\u00e9tico existen dos l\u00edneas principales: la linfoide, que da lugar a las c\u00e9lulas T, los linfocitos B y las c\u00e9lulas NK, y la mieloide que formar\u00e1 los eritrocitos (gl\u00f3bulos rojos) o los macr\u00f3fagos entre otros.\u2028Seg\u00fan explic\u00f3 Maribel Parra, para que se den estos procesos de diferenciaci\u00f3n deben producirse una serie de cambios en el programa g\u00e9nico de las c\u00e9lulas. Existen factores de transcripci\u00f3n que activan genes de cada linaje celular pero tambi\u00e9n existen represores transcripcionales que impiden que se expresen genes “inapropiados”, que son caracter\u00edsticos de otros tipos celulares.\u2028\u2028El grupo de Maribel Parra estudia precisamente el papel de una familia de prote\u00ednas que son represoras transcripcionales en la diferenciaci\u00f3n de las c\u00e9lulas del sistema hematopoy\u00e9tico. Se trata de las histonas desacetilasas (HDAC).

En este trabajo los investigadores demuestran que el papel de una de estas prote\u00ednas, la HDAC7, en linfocitos B es reprimir genes \u201cinapropiados\u201d, caracter\u00edsticos de otros tipos celulares como los macr\u00f3fagos y los linfocitos T. Los investigadores han utilizado un modelo de rat\u00f3n deficiente en HDAC7 en progenitores de c\u00e9lulas B (c\u00e9lulas pro-B) y han observado que HDAC7 es esencial para el desarrollo de linfocitos B. Los investigadores demuestran que en c\u00e9lulas pro-B HDAC7 se une a dichos genes \u201cinapropiados\u201d resultando en su represi\u00f3n transcripcional. Maribel Parra ha explicado que “a trav\u00e9s de este mecanismo de represi\u00f3n transcripcional, la c\u00e9lula B se asegura de que no se expresen genes de otros tipos celulares, manteniendo su identidad linfoide”. Tambi\u00e9n se\u00f1ala que “dada su importancia en la biolog\u00eda de los linfocitos B, la funci\u00f3n de HDAC7 podr\u00eda estar alterada en enfermedades hematol\u00f3gicas. De hecho el a\u00f1o pasado publicamos un art\u00edculo en el que mostr\u00e1bamos que la expresi\u00f3n de HDAC7 est\u00e1 desregulada en tipos espec\u00edficos de leucemias y linfomas de c\u00e9lulas B.\u201d
